Objective:Colorectal cancer is a common cancer in developed countries such as North America, Europe and other countries. Colorectal cancer is often the common visceral cancer, with the incidence rate of up to 35-50/10 million. From 2000 to 2002, in China's 27 provinces, autonomous regions and municipalities, cancer mortality survey (about 10% of the population over the same period) found that colorectal cancer incidence rate of 4 per cent annual growth rate increased rapidly, accounting for gastrointestinal The second tumor. Colorectal cancer can occur early lymph node metastasis, is the main reason for clinical treatment failure. At present, tumor metastasis of cancer-related genes was subject to the regulation. Research shows that at least a dozen of the cancer genes that increase or decrease can be induced or to promote the transfer of potential cancer cells, such as tumor apoptosis gene bcl-2, bax, p53, Fas / FasL, such as tumor suppressor gene transfer nm23, KAII , KISSI, MKK4, BrMSI, etc.Nm23 gene transfer, as a tumor suppressor gene in tumor metastasis conditioning control of the many forms of "transfer suppressor gene", is the most eye-catching. In recent years two people have been isolated from the nm23DNA cloning, that is nm23-H1 and nm23-H2 type. In some high Nm23 gene transfer of tumor cells, can be expressed in decline, allelic loss and gene mutation. Fas antigen (AP0-1/CD95) and Fas ligand (FasL) composed of Fas system, will turn into apoptosis signal cells play an important role. FasL combined effect of Fas antigen target cells, within a few hours by apoptosis ways targeted cell death. In recent years many studies found that Fas in a variety of cell surface expression of FasL expression and only a small number of cell types, such as lymphocytes, immune cells and privileges of multiple organ cancer cells. In a variety of tumor cells expression of Fas and FasL expression increases down the phenomenon, with the removal of this tumor-infiltrating lymphocytes and curb anti-tumor immune responses, involved in the immune escape. This study used the immunohistochemistry to detect nm23, Fas / FasL in colorectal cancer tissues and explore its local infiltration and the relationship between metastasis and clinical significance.Materials and Methods:1. Experiments Materials(1)The source of Samples51 cases of colorectal cancer samples are in Liaoning Province Cancer Hospital of surgical resection of colorectal cancer archive wax block from November 2007 to 2008 February. Men ,33 cases; 18 cases of women aged 26-81 years, with an average age of 55.34, tumor site: 33 cases of rectum, sigmoid seven cases, five cases of transverse colon, colon or six cases; general types: 31 cases of ulcers, uplift of 20 cases ; Organizations type: high score of 20 cases, 28 cases in the division, poorly differentiated three cases of clinical pathology in phases: Dukes'A period of five cases, Dukes'B period of 19 cases, Dukes'C period of 13 cases, Dukes'D period of 14 Cases. All the cases have been in before colonoscopy, conventional pathological examination for cancer diagnosis. Cancer and more cases of cancer recurrence are ruled out. Patients before did not receive radiotherapy and chemotherapy and other treatment for other tumors. All pathological specimens of colorectal cancer by 10 percent of the neutral formalin are fixed, embedded in paraffin and sliced. And nine cases from adjacent tumor from 5cm above the normal colon tissue and 44 cases with fiber colonoscopy removal of specimens for colorectal adenomas-control study.(2)Experimental reagentsReagents nm23,Fas / FasL mouse anti-human monoclonal antibody and SP kit, are all the reagents from Fu jian new reagent products.2. Experimental MethodsImmunohistochemistry: Streptomycin uses conventional antibiotics protein–peroxides connection (Streptavidin-Peroxidase,SP) immunohisto- chemical SP method to control anti-human nm23,Fas / FasL-used as a monoclonal antibody anti-cancer detection nm23 and Fas / FasL Protein expression.Nm23 protein expressed in the form of the cytoplasm in brown granules for the positive cells, Fas / FasL protein in cell membranes and (or) cytoplasm appear brown granules for positive cells. AB semi-quantitative method score results.3. Statistical analysisSPSS11.5 used statistical analysis software andχ2 test. And the two-factor analysis of the correlation usedχ2 test and both P <0.05 significant.Results:(1)Colorectal cancer and normal epithelial tissue nm23,Fas / FasL expressionColorectal cancer nm23, Fas expression of the lowest, followed by colorectal adenomas, is the highest expression of normal colon; FasL in different colon, the expression of Fas the contrary and the expression of colorectal cancer in the highest are followed by the colon Adenoma, in almost no normal expression of FasL. Each group nm23﹑ Fas﹑ FasL expression differences were statistically significant.(2) Nm23,Fas / FasL expression and colorectal cancer clinical and pathological features of the relationshipStatistics show nm23,Fas / FasL expression and gender in patients with colorectal cancer tumor site age has nothing to do with the degree of differentiation, the depth of invasion, metastasis and the Dukes' stage.(3) Colorectal nm23 organizations and the coordination of Fas expression Nm23 and Fas expression in colorectal cancer are in related relations.Conclusion:(1)The expressions of Nm23,Fas / FasL system protein in colorectal cancer tissues and in normal colon and colon adenoma show a significant difference;(2)Nm23,Fas / FasL protein expression and colorectal cancer patients with age and gender of the tumor site has nothing to do with the tumor differentiation, depth of invasion, metastasis and Dukes' staging related;(3)Nm23 and Fas expression in colorectal cancer are in related relations.
