| Objective: To investigate the expression of ING4(inhibitor of growth family member 4),P53,Ki-67,Fas/FasL in bladder urothelial carcinoma,and explore the expressions of ING4,P53,Ki-67 and Fas/FasL in the role of development of bladder urothelial carcinoma and clinical significance,and analysis the relationship between ING4 and P53,Ki-67,Fas/FasL,as well as providing theoretical basis for biological marker,gen thearpy of bladder cancer,early diagnosis,post-operation and prognosis judgment of bladder cancer.Methods: The expressions of ING4,P53,and Ki-67,Fas/FasL was detected by immunohistochemical staining assay of 60 patients of bladder urothelial carcinoma,60 samples of corresponding cancer adjacent tissues and37 samples of normal bladder mucous membrane tissues from North Sichuan medical college affiliated hospital and Nanchon central hospitalin the period from 2014 to 2015 by immunohistochemical staining with ElivisionTM Plus two-step method.Result:1.The positive expression rate of ING4 in urothelial carcinoma,tissue adjacent to carcinoma and normal bladder mucous membrane tissues respectively was 70%(42/60),100%(60/60)and 97.2%(36/37),with signifacant difference(P<0.05).tissue adjacent to carcinoma and normal bladder mucous membrane tissues respectively had no signifacant difference(P>0.05).The positive expression rate of ING4 in advanced level or muscle invasive is lower than low grade or non invasive(P<0.05).2.The positive expression rate of P53 in urothelial carcinoma,tissue adjacent to carcinoma and normal bladder mucous membrane tissues respectively was 96.7%(58/60),13.3%(8/60)and 13.5%(5/37),with signifacant difference(P<0.05).Tissue adjacent to carcinoma and normal bladder mucous membrane tissues had no signifacant difference(P>0.05).The positive expression rate of P53 in advance level or muscle invasive is higher than low grade or non invasive(P<0.05).3.The positive expression rate of P53 in urothelial carcinoma,tissue adjacent to carcinoma and normal bladder mucous membrane tissues respectively was 91.7%(55/60),11.7%(7/60)and 2.7%(1/37),with signifacant difference(P<0.05).Tissue adjacent to carcinoma and normal bladder mucous membrane tissues had no signifacant difference(P>0.05).The positive expression rate of Ki-67 in advance level or muscle invasive is higher than low grade or non invasive(P<0.05).4.The positive expression rate of Fas in urothelial carcinoma,tissue,adjacent to carcinoma and normal bladder mucous membrane tissues respectively was 70%(42/60),88.3%(53/60)and89.2%(34/37),with signifacant difference(P<0.05).Tissue adjacent to carcinoma and normal bladder mucous membrane tissues had no signifacant difference(P>0.05).The positive expression rate of Fas in advance level is higher than low grade(P<0.05).5.The positive expression rate of FasL in urothelial carcinoma,tissue adjacent to carcinoma and normal bladder mucous membrane tissues respectively was 76.7%(46/60),100%(60/60)and94.6%(35/37)with signifacant difference(P<0.05).tissue adjacent to carcinoma and normal bladder mucous membrane tissues had no signifacant difference(P>0.05).6.To compare and analyze expression of ING4,P53,Ki-67 and Fas/FasL in the bladder cancer by using Spearman rank correlation,ING4 showednegative correlation between P53,Ki-67.ING4 was positively related to Fas/FasL.Conclusions1.ING4 was low expression or deletion in bladder cancer tissues,suggesting that it has been tumor suppressor genes in the pathogenesis of bladder cancer.With inceased clincal stage or pathologic grade of bladder cancer,the expression was decline,suggesting that it has been close relationship between with tumor invasive.therefore ING4 is a good candidate as a clinically relevant indicator of disease progression2.P53 was high expression in bladder cancer tissues,and as well as with inceased clincal stage or pathologic grade and tumorous numberof bladder cancer,suggesting that the excessive expression of P53 protein is one of the symbols of highly malignant bladder cancer.3.Ki-67 was high expression in bladder cancer tissues,and as well as with inceased clincal stage or pathologic grade and tumorous numberof bladder cancer,suggesting that Ki-67 can be a good indicator of bladder cancer pathogenesis,diagnosis and prognosis,and as important clinical significance ind predicting early recurrence,deterioration and therapeutic effect.4.Fas was low expression or deletion in bladder cancer tissues,suggesting that bladder cancer may have pathways resistant to Fas-mediated autocrine cell suicide.Thus,the loss of Fas was suggested to be a crucial factor for selecting patients requiring more aggressive treatment.5.The rate expression of FasL in bladder cancer was lower than benign lesion,suggesting that FasL played an important role in bladder cancer with benign lesion becomed malignancy and didn’t change along with the degree of cell differentiation,growth.The rate expression of FasL in normal tissue cancers is higher than bladder cancer,requiresed further study.6.ING4 showed negative correlation between P53,Ki-67 and was positively related to Fas/FasL,suggesting that the occurrence and development of bladder cancer was a complex process of factors and gene.Finally combined detection of expression may help for the biological marker,clinical diagnosis,treatment and prognosis of bladder urothelial carcinoma. |
