| Objective To investigate the significance of the expression of vascular endothelial growth factor (VEGF), its receptor KDR and FLT-1 in brain astrocytomas. Methods The expression of VEGF, KDR and FLT-I was examined immunohistochemistrically by PV-9000 method in 47 cases astrocytomas . Results There was an extensive expression of VEGF ,KDR and FLT-1 ,mainly in the cytoplasm of tumor cells (TC) and endothelial cells (EC) .The lebeling index (LI) of VEGF ,KDR and FLT-1 in TC was 26.61±2.40, 22.8fttl.76, 30.80±2.56; in EC was 18.82±1.65, 50.40±3.31, 29.34±2.68 , respectively the LI of VEGF in TC was higher than that in EC (F=7.124, P<0.01) .The LI of KDR in EC was higher than that in TC (F=53.74, P<0.01) , and the expression of FLT-1 in TC and EC was found at similar levels (F=0.155, P>0.05) The expression of VEGF ,KDR and FLT-1 ,in TC and EC were all significant among three groups with varying gradings ( F=26.4, 13.4, 5.2, 14.3, 16.4, 10.9; P<0.01 , 0.01, 0.01, 0.01, 0.01, 0.01) .The expression of VEGF in TC was correlated with that of KDR and FLT-1 ( r=0.649, 0.864; P<0.01, 0.01 ) .Expression of VEGF in TC was correlated with that of KDR and FLT-1 in EC (i=0.831, 0.717; PO.01, 0.01). Expression of VEGF in the EC was correlated with that of KDR and FLT-1 (r=0.762, 0.633; P<0.01, 0.01) . Expression of VEGF in EC was correlated with that of KDR and FLT-1 in TC(r=0.615, 0.702; P<0.01, 0.01).Conclusion K VEGF and the receptor KDR ,FLT-1 were all expressed not only in TC but also in EC .VEGF was mainly expressed in TC ,KDR was mainly in EC, There was no difference betwent expression of FLT-1.2, The expression of VEGF and the receptor KDR and FLT-1 was all related to varying pathological grades .3, The growth of TC and EC was both enhanced by autocrine and paracrine. TC was mainly in autocrine and EC was mainly in paracrine. 4, Detecting the expression of VEGF , KDR and FLT-1 in brain astrocytomas may be favour to instructing personal treatment and evaluating prognosis. |
