Objective To explore the expression of anti-oncogene P53 , PTEN(phosphatae and tension homologue deleted on chromosomelen ) and MMP-9 (matrix metalloproteinases 9) in the hepatocellular carcinoma, and the relation of them and clinical pathology and the prognosis. Methods Forty-one specimens of HCCs, who were followed up, were obtained from the operative specimens from the Affiliated Hospital of Qingdao University Medical College. The P53, PTEN and MMP-9 were measured by the immunohistochemistry two-step methods of PV-6000. Results The five years survival rate of overall patients is 44.4%, media survival time is 42.34 months, Small HCC (diamater≤5cm ) is 66.9%, big HCC(diamater>5cm ) is 28.2 % respectively. The positive expression rate of P53, PTEN and MMP-9 is 63.4%, 48.8% and 73.1% respectively. The five years survival rate for each positive expression group is 36%, 52.3% and 36.9%. By spearman relative analysis, we found that the positive expression of P53 and PTEN have significant correlation (r=0.374, P=0.045),and every positive expression of them also is relative to blood AFP concentration (r=0.404, P=0.033 and r =0.46, P=0.014) and Edmondson differ degree(r=0.350, P=0.046 and r=0.377, P= 0.036).The positive expression of MMP-9 is also relative to Edmondson differ degree(r= 0.404, P = 0.0204). In both monovariate Kaplan-Meier Plot and multivariate Cox regression analysis, the positive expression of P53, PTEN and MMP-9 were not linked to unfavorable prognosis. Conclusion (1) P53 is high expression in HCC tissue and represent the variety expression in the difference progressive phases of HCC; (2) PTEN is absent expression in HCC tissue and maybe assist the progress of HCC tissue differ; (3) MMP-9 is high expression in HCC tissue and maybe take part in the deteriorate progress of liver tissue; (4) P53, PTEN and MMP-9 maybe join the pathological change progress of HCC;(5)Those patients have a better prognosis that have the negative expression of P53 and MMP-9 and positive expression of PTEN together.
