Squid, one of the most important cephalopod marine creatures in the coastal water of our country, is rich in nutrition and has a variety of medicinal functions.From early 1990s, Japanese scholars found that squid ink had high anti-tumor activity. Most domestic researches are focused on the anti-tumor activity of squid ink homogenate. Fresh squid ink was used in our present study as the experimental material, which was added with a proper amount of aseptic normal saline to be homogenized. Then, it was degreased by using acetone at a temperature of -20℃, centrifuged, precipitated, freeze-dried and reacted with 0.1 M Tris-HCl buffer solution (pH6.8) for 72 hours at a temperature of 4℃. Then, once again, it was centrifuged and the supernatant fluid was taken from the preparation, which was dialyzed at 4°C in the distilled water for 48 hours. After that, the treated preparation was concentrated, freeze-dried and the final extract of the squid ink peptidoglycan (SIPG) was obtained. The extract SIPG was dissolved in 0.1 M Tris-HCl buffer solution, passed through the DEAE-Sephacl to have ion-exchange chromatography, then it was washed step by step with 0, 0.1, 0.2, 0.3, 0.4, and 2.0 M concentration of NaCl and the washed fluid was collected and desalted by means of...
