Epimedium koreanum is a traditional Chinese medicine, which is included inChina Pharmacopoeia and grows in the Changbai Mountain area. Lots of experimentsshow that Herba Epimedii has the activity of enhancement of immunity, anti-aging,anti-inflmmatory, anti-arrhythmia and anti-microbial. This paper mainly studies on theseparation and purification of derivative I of flavonoids and the evaluation ofanti-fatigue and anti-oxidant activity.1. The separation and purification of derivative I of flavonoidsThe leaves of Epimedium koreanum were smashed and extracted twice with70%ethanol solution. In acidic conditions, derivative I of flavonoids was prepared. Thecrude extract was purified by gradient pH extraction method. Using high performanceliquid chromatography, the content of the purified product was determined. The resultshowed that the content of derivative I of flavonoids was up to75%.2. The methodological study of derivative I of flavonoids by HPLCIn this paper, the high performance liquid chromatography was used todetermined derivative I of flavonoids, gradient elution, mobile phase of acetonitrile (A)-water (B) mixed solvent; detection wavelength of272nm, the column temperaturewas30℃, the flow rate was1.0mL/min conditions. The derivative I of flavonoidswas better separated.The results showed that the linear range of derivative I of flavonoids was6.14μg/mL~92.1μg/mL and the accuracy and the precision were well. The minimum detection limit was0.184μg/mL; the lowest detection limit was0.614μg/mL; thespecificity, durability, stability and reproducibility were good. This is a good way toconduct quality control which can be applied to determine the derivative I offlavonoids content.3. The evaluation of anti-fatigue and anti-oxidant activityIn this paper, the motion test and biochemical parameters were combined toevaluate the anti-fatigue and anti-oxidant activity of derivative I of flavonoids. Theswimming time, blood lactic acid (LD), blood urea nitrogen (BUN), liver glycogen,hepatic malondialdehyde (MDA) and SOD activity were determined.The results showed that derivative I of flavonoids could significantly prolong theswimming time, decrease content of blood lactic acid (LD) and blood urea nitrogen(BUN), increase the reserves of liver glycogen and the SOD activity, and speed up therate of malondialdehyde. Derivative I of flavonoids has remarkable activity ofanti-oxidation and anti-fatigue.The results of this study laid a foundation of further pharmacodynamic study andhealth products.
