Objective: To investigate the expression of MMP-3 JMMP-13 and CD44V5 proteins in tissues of gastric cancer and explore the mechanism of invasion and metastasis in gastric cancer. Methods: 60 paraffin-embedded samples of gastric cancer and para-cancer from surgically resected tissues were selected, 15 biopsied endoscopically samples of chronic surface gastritis were served as controls. The expression of MMP-3, MMP-13 and CD44V5 proteins were examined by immunohistochemical method. Results: The positive expression rates of MMP-3, MMP-13 and CD44V5 proteins in samples from cancerous tissue were 60.6%, 68.3% and 48.3% respectively, but none of these proteins were detected in control tissues. Those of MMP-3, MMP-13, CD44V5 proteins in matched para-cancer tissues were 16.7%, 21.7%, 10.0% respectively. The positive expression rates of cancerous tissue were significantly higher than those of para-cancer tissue (P < 0.05) . The expression of MMP-3, MMP-13 and CD44V5 proteins correlated statistically with the lymph node metastasis and cancer stage. There were high significant relationship among the MMP-3, MMP-13 and CD44V5 protein expression(P < 0.05). The lymph node metastasis rate of tissues with positive expression of two or more parameters was significantly higher than tissues with positive expression of only one or none parameter(P < 0.05). Conclusions: The expression of MMP-3 ,MMP-13 and CD44V5 are correlated to invasion and metastasis of gastric cancer ,and thus could be used as markers to predict metastasis and invasion of gastric cancer and the prognosis of these patients as well.
