Objective:To study the expression of EphA2, c-Met and CD44v5 in esophageal squamous cell carcinoma(ESCC) tissue and corresponding adjacent non-carcinomatous tissues, and explore its relationship with clinicopathological significance in Kazakhand Han ESCC.Methods:To detect the EphA2,c-met and CD44v5 mRNA and protein in Kazakh and Han ESCC tissues and its corresponding tissues by RT-PCR and immunohistochemical methods(IHC); and use Western-Blot and enzyme-linked immunosorbent assay (ELISA) to detect the protein in serum of ESCC patientsand healthy cases.Rusults:(1)The IA value of EphA2mRNA in carcinoma tissue (0.488±0.141) is higher than corresponding adjacent tissues (0.137±0.127) with statistically significant(P<0.01);And the positive expression of EphA2 protein in carcinoma tissue is higher thancorresponding adjacent tissues (P<0.05);The expression of EphA2 is related to invasive depth,TNM stages and lymph node metastasis in tissues(P<0.05).The level of EphA2 protein in serum of ESCC patients is higher than the healthy controls(P<0.01), and the value of this protein in serum of ESCC have relationship with invasive depth and TNM stages(P<0.05).(2)c-met:The IA value of c-metmRNA in carcinoma tissue (0.748±0.122) is higher than corresponding tissues(0.147±0.064) with statistically significant(P<0.01);and the positive expression of c-met protein in carcinoma tissues is higher than corresponding tissues(P<0.01);The expression of c-met is related to invasive depth,TNM stages and lymph node metastasis in tissues(P<0.01).The level of c-met protein in serum of ESCC patient is higher than thehealthy controls(P<0.01), and the value of this protein in serumhave relationship with invasive depth, lymph node metastasis and TNM stages(P<0.05).(3) CD44v5:The IA value of CD44v5mRNA in carcinoma tissues (0.737±0.116) is higher than corresponding tissues (0.140±0.065) with statistically significant(P<0.01);and the positive expression of CD44v5 protein in carcinoma tissue is higher than corresponding tissues(P<0.01);The expression of CD44v5 is related to differentiation,TNM stages and lymph node metastasis in tissues(.P<0.01).The level of the protein in serum ofESCC patients is higher than the value of healthy controls(P<0.01), and the value of this protein in serum of ESCC have relationship with differentiation, invasive depth, lymph node metastasis and TNM stages(P<0.05).(4) There’s no statistic significant difference between Kazakh and Han of the expression of EphA2,c-met and CD44v5(P>0.05).There is correlation between the expression of c-met and CD44v5 (r=0.960, P<0.01).Conclusion:The expression of EphA2, c-met and CD44v5 was related to the occurrence,invasion and metastasisof ESCC both in Kazakh and Han.EphA2,c-met and CD44v5 protein in serum may be reminders of the occurrence, early diagnosis and evaluation indicator of ESCC.C-met can regulates the expression of CD44v5 via some methods perhaps.
