The Effect Of Defibrase On Laminin And IV Collagen In Traumatic Rabbit Eyes With Proliferative Vitreoretinopathy

OBJECTIVE To study the effect of Defibrase on Laminin and IVcollagen in experimental traumatic proliferative vitreoretinopathy.METHODS Our experiment introduce the experimental model induced by posterior penetrating eye injury and vitreous infusing blood with or without defibrase in the twenty rabbits. These rabbits were divided into five groups by random: normal control, physiological saline group, full blood group, and defibrase group which was subdivided into 0.03u group and 0.06u group according to the defibrase concentration. Five eyes in the normal control and physiological saline group. Ten eyes in the full blood group , and defibrase group. Anesthesia by intramuscular injection at rabbit stern. Then at the temple side 1:00 aspect, 3mm after the limbus of cornea, we injected toward the certer of ball perpendicularly throught pupil under euthyphoria with 7# needle, extracted 0.3ml vitreous body. In physiological saline group, we injected 0.3ml physiological saline into the vitreous cavity .In full blood group, we injected 0.3ml blood into the vitreous cavity through the auricle vein. In defibrase 0.03u group, we injected 0.3ml blood extracted from auricle vein and 0.1ml 0.03u defibrase. In defibrase 0.06u group, we injected 0.3ml blood extracted from auricle vein and 0.1ml 0.06u defibrase. Don't sut the cut, then droped 0.25% chloroptic into conjunctival sac, intramuscular injection penicillin( 80 million unit) at rabbit stern. Executed all the rabbits after 40 days and enucleated the eyes immediately. Injected 10% formalin into vitreous body, then soaked into 10% formalin liquid a week to fast the sample. A week later, cut open the eyes at the level of cornea and optic nerve, then alcohol dehydration, wax immerses, paraffin embedding, and then dyed the chip with HE and immunohistochemical SP routinely. With DMR+Q550 pathological image analysator, counted the positive dyeing area ratios (the positive dyeing areas of a visual field compares with the total areas of a visual field) with high power lens.
