A lot of oculopathy can induce vitreoretinal proliferative membranes and traction-band, which can cause retinal detachment and other sever ocular diseases. If these patients don't accept prompt ly therapy, whose vision acuity will be badly threatened.Proliferative vitreoretionpathy (PVR) is a severe blind_causing ophthalmopathy, and it is the main cause of the failure of retinal detachment surgery. Traumatic PVR is the end of fibrous tissue's over proliferation after perforating injury of eyeball. Proliferative diabetic retinopathy (PDR) is the complication of long-term hyperglykemia. The chang of membranes's composition deduce retina ischemis,and blood suger's abnormal raise can also deduce the functional disorder of capillary pericyte and the functional injury of blood—retina barrier. There are many difference between these three eye diseases, more over there are some common performance, e.g.,the destruction of blood—reina barrier; the exposure and migration of retinal pigment epithelial, the exudation of cells factor, the inducing and chemotacting of some kinds of cells's infiltration e.g., retinal glial cells,macrophage and fibroblast. All these cells produce lots of cellular factor, and form proliferative membrance in the end. The contraction of these proliferative membrance cause a serious ocular complication, whose main performance is retinal detachment.The patient's visual acuity and everyday life are badly impacted by these serious complications.We use five species specific anti—bodys to investigate cell type in different proliferative membranes , because of immunohistochemical staining's character.And then analyzeing these cells roles in PVR,Traumatic PVR及PDR..Objective:The experiment was arranged to investigate and compare the cellular composition in the proliferative membrance of PVR,traumatic PVR and PDR by immunohistochmeical staining. The similarities and differences of the cellular composition in proliferative membrances are also approached in this experiment. Using the result of this experiment, we could offer some ground of treatment.Method:The specimens was obtained by performing vitrectomy from patiences with PVR(16 cases),traumatic PVR(12 cases),PDR(7 cases). Grouping all cases into three groups, named A group, B group, C group. Fixation them with 0.1%GA, and then embedded them into olefin. The next step was to shear every specimen into six pieces, which was used to be stained. And then, these stained specimens were readed and taken pictures under microscope. Finally, using statistics to analysed the result.Result: The results of HE staining: Every specimens was composed of cellular components and extracellular matrix. The composion of cellular components was RPE cells,fibroblast,retinal glial cells and macrophage. All cells were surrounded by extracellular matrix. During the course of diseases, the cell's proportion in these specimens was decreasing and extracellular matrix's proportion is increasing. Every kind of cell's proportion was always changing. Compared with PVR and traumatic PVR specimens, PDR specimens had lots of neovascular tissues, besides retinal pigment epithelium,fibroblast,retinal glial cells and macrophage.In all 16 PVR specimens, positive staining was found in 15 for CK. 16 for GFAP and FN. But it changed in the course of disease. Parts of specimenswere stained CD68,FN and FⅧpositive .The traumatic PVR group had 11,8 and 9 specimens stained CK,GFAP and CD68 positive. All traumatic proliferative membrances's cellual members were decreasing and extracellular matrix were increasing in the couse of disease.All PDR specimens were stained FⅧpositively. This result manifested lots of vascular tissues. Other cellular elements were partly stained positively.Conclusion:(1)All proliferative membrances in three groups basicly has the same cellular elements, but every kinds of cells doesn't take the same proportion in three groups. (2)Retinal pigment epithelium,fibroblast and retinal glial cells take the main cellual elements in PVR and traumatic PVR specimens, and vascular structure take the main elements in PDR specimens.(3)Every cells'proportion in each groups is changing in the couse of disease differently.
