Objective: Complex of HLA is the inherit system that is related to the disease discovered at first. It is the human immunity genetics system that is investigated more than others. Recently many investigators have searched for an association between certain HLA class I or II genotyping and chronic HBV infection . But conflicting data were obtained from these studies. To investigate the association between different outcomes of HBV infection and special inherit background, discuss the mechanism of chronic hepatitis B and the effect of HLA genotyping on hepatitis B.We selected HLA-B,C to our study objects ,estimated the genotyping of HLA-B,C to patients with chronic hepatitis B and healthy objects of the Han nationality in Shandong district by PCR-SSO ,analysis difference of genotyping frequency of HLA-B,C between patients and healthy objects and search the genotyping of HLA -B,C that is related to hepatitis B. We want to provide theory foundation to forecast, prevent and therapy hepatitis B.Methods: 29 hepatitis B patients from shandong of the Han nationality were collected 2ml fresh heporinized blood. HLA-B,C genotyping was conducted in all patients with polymerase chain reaction -Sequence Specific Oligonucleotide, and compared with healthy subjects from shandong of the Han nationality. The relationship of different genotypes of HLA-B,C with the clinical types of HBV infection was analyzed.Results: HLA-B55, HLA-B 71, HLA-B 72 and HLA-C3 were frequent genotypes in the patients with chronic hepatitis B from shandong of the Hannationality than in the healthy residents. The frequency of HLA-B54 was decreased in the patients with chronic hepatitis B compared with healthy subjects. The difference of genotypes frequency of HLA-C3,HLA-B55,71,72,54 between the patients and the healthy subjects reached statistical significance(/><0.05). HLA-C15 was frequent genotypes in the patients with cirrhosis than in the non-cirrhosis patients(/><0.05).Conclusion: HLA-B55/71, 72 and HLA-C3 were chronic hepatitis B of the Han nationality in Shandong district. And HLA-B54 was related to anti -infection of hepatitis B virus. HLA-C15 was related to cirrhosis. Immune and genetic factors participate in the pathogenesis of hepatitis B.
