[Objective]To study the expression of HIF-1α and VEGF in gastric cancer and their relationship with tumor angiogenesis, clinicopathologic feature and prognosis. [Methods]Immunohistochemical technique was used to detect the expression of HIF-1α and VEGF in 80 cases of gastric cancer, the microvessel density(MVD) marked by CD34 was also detected. The relationship between the expression of HIF-1α , VEGF and tumor angiogenesis, clinicopathologic feature, prognosis of patients was then analyzed. 30 cases of gastric by-cancer tissue and 30 cases of normal gastric tissue were selected as contrast group. [Results]The MVD of gastric cancer(30.2±6.3) was significantly higher than that of gastric by-cancer tissue (13.1 + 1.6) and normal gastric 1 tissue (6.5 + 2.7), P<0.01. The MVD of gastric cancer was significantly correlated with TNM stage, invasive depth and lymph-node metastasis, P<0.01.In the cases of gastric cancer ,the 5 years survival rate from the Kaplan-Meier survival analysis in the high MVD group(21.9%) was significantly lower than that in the low MVD group(42.9%), Logrank=6.24, P =0.0125.The positive expression rate HIF-1α in gastric cancers(47.5%) was remarkably higher than that gastric by-cancer tissue (20.0%) and normal gastric tissues (0%) , P<0.05. In gastric cancer, the expression of HIF-1α was remarkably associated with TNM stage, differentiated degree, invasive depth and lymph-node metastasis, P<0.05. The MVD of the HIF-1α positive expression group (33.6 ± 6.6) was remarkably higher than that of the negative expression group (27.1±4.1 ), P =0.000; the positive expression rate of HIF-1 a in high MVD group(59.1%) was remarkably higher than that in low MVD group(33.3%), P<0.05. In the cases ofgastricl cancer, the 5 years survival rate from the Kaplan-Meier survival analysis in the HIF-1 a positive expression group(15.2%) was remarkably lower than that in the negative expression group (50.0%), Logrank=15.55, P=0.0001.The positive expression rate of VEGF in gastric cancers(71.3%) was significantly higher than that in gastric by-cancer tissue (43.3%) and normal gastric tissues (10.0%), P<0.05. In gastric cancer, the expression of VEGF was significantly correlated with TNM stage, invasive depth, lymph-node metastasis and tumour size , P<0.05. The MVD of the VEGF positive expression group (31.2 + 6.3) was significantly higher than that of the negative expression group( 27.5 ±5.5 ), P=0.016; the positive expression rate of VEGF in high MVD group(83.7%) was significantly higher than that in low MVD group(51.6%), P=0.005. In the cases of gastric cancer ,the 5 years survival rate from the Kaplan-Meier survival analysis in the VEGF positive expression group(22.9%) was significantly lower than that in the negative expression group (57.9%), Logrank=8.51, P=0.004.In the cases of gastric cancer, the MVD of both HIF-1 a and VEGF positively expressed group, one of them positively expressed group, both of them negatively expressed group were 34.3±5.4, 28.9±4.6, 24.8±5.3 respectively, P<0.01; There was a positive correlation between expression of HIF-1 a and VEGF, Y =0.328, P= 0.003. [Conclusion]1. The overexpression of HIF-1 a and VEGF in gastric cancers have a significant relation with tumor angiogenesis.2. HIF-1 a , VEGF and MVD are significantly correlated with TNM stage, invasive depth and lymph-node metastasis of gastric cancer, and VEGF is correlated with tumour size; their overexpression may lead to advanced invasion of gastric cancer and unfavorable prognosis.3. HIF-1 a and VEGF may have play a corporative action in gastric cancer, their simultaneous expression may lead to tumor angiogenesis increasing greatly, cancer invading and metastasizing easily.4. The combinative detection of HIF-1 a and VEGF in gastric cancer have the positive significance of the judgement of malignant phenotype and prognosis, and guide to the clinical treatment.
