| Background: Cervical cancer is the most frequent malignant disease infemale genital system. The development of cervical cancer is closely associated with the presence of certain human papillomavirus types such as HPV-16, 18. A polymorphism was found in codon 72 of human p53. There may be a correlation between p53 codon 72 polymorphism with HPV 16, 18 E6 and cervical cancer. Anyway, the p53 codon 72 polymorphism may be a potential marker of cervical cancer.Purpose: To analysis the relationship between p53 codon 72 polymorphismand cervical cancer, HPV16/18 E6.Methods: 81 cervical squamous cancer, 18 cervical adenocarcinoma, 88CIN â…¡~â…¢ and 60 normal cervical specimens were included in this study. PCR was used to examine p53 genotypes and HPV16, 18 E6.Results:1) The frequency of p53 Arg homozygosity in cervical squamous cancer cervical adenocarcinoma and CIN â…¡~â…¢ was 58.02% , 55.55% and 59.09% respectively, which is much higher than that of p53 Arg/Pro heterozygosity and of p53 Pro homozygosity in each groups, and higher thanthe frequency of p53 Arg homozygosity in normal samples.2) There is no statistic difference in normal samples for the frequency of p53 Arg homozygosity , p53 Arg/Pro heterozygosity and p53 Pro homozygosity.3) The frequency of HPV16, 18 E6-positive in cervical cancer and CIN was much higher than that in control group(81.84%, 50.00% and 53.41% versus 15.00%). Compared with CIN, the frequency of HPV16 , 18 E6-positive was higher in cervical squamous cancer.4) The frequency of p53 Arg homozygosity in HRHPV E6-positive cervical squamous cancer samples is much higher than that in HRHPV E6-negative cervical squamous cancer samples and in HRHPV E6-positive normal samples.5) The allele frequency of p53 Arg is much higher than that of p53 Pro in malignant and premalignant groups.6) No difference of p53 codon 72 polymorphism was found according to FIGO stagings and grades.Conclusion:Based on the findings of this study, it is suggested that p53 Arg homozygosity could act as a potential risk factor for the tumorigenesis of the cervix. p53 codon 72 polymorphism has no relation with the FIGO stagings and grades of cervical cancer. p53 Arg homozygosity and HRHPV E6-positive simultaneously can predict the fate of cervical lesions. |
