Part oneRadix rehmanniae is one of beneficial traditional Chinese medicine. Study on the literatures of iridoid glucosides from R. rehmanniae show that catalpol has hypoglycemic effect on alloxan-induced diabetic mice. Iridoid glucosides from R. rehmanniae have large polarity, similar structure and poor stability. On the grounds of it, We make the study on the extraction process for catalpol in R. rehmanniae by orthogonal design. The best extraction process is adding 4 time amount of 50% alcohol into the R. rehmanniae and extraction 3 times, 2 hours for each time. Experimental results obtained from active carbon and macroporous absorption resin column showed that the adsorption capacity of active carbon is higher than macroporous absorption resin. The result of determination of active substance catalpol show that: when intraperitoneal injection and subcutaneous injection, catalpol has hypoglycemic effect on Alloxan-induced Diabetic Mice. Catalpol has no inhibition effect of a-glucosidase and aldose reductase.Part twoEpimedium wushanense mainly distributes in Shaanxi province, Sichuan province and Guizhou province . In national pharmacopoeia of 2000 edition, E wushanense, along with another four herbas such as E. brevicomwn, E. sagittatum, E. pubescens and E. koreanum were all recorded as herba epimedii. There are not many published paper reaching in their chemical constituents at present.The components were separated on polyamide and silica gel column chromatography and the structure was identified by spectral evidence of UV,IR, NMR and MS.Three compounds, which are icarisid I , rouhuoside and icariin, were isolated from E. wushanense root. We establish a HPLC method to determine the content ofIcariside I ,rouhuoside and icariin in E Wushanense. We determined nine samples by this HPLC method.After all the work, we are successfully complete the task of People's Republic of China Pharmacopoeia of 2005.
