Epimedium wushanense mainly distributes in shaanxi province, Sichuan province and Guizhou province. In national pharmacopoeia of 2005 edition, E. wushanense, along with another four herbas such as E. brevicornu Maxim, E. sagittatum (Sieb.etZucc.) Maxim, E. pubescens Maxim, E. koreanum were all recorded as herba epimedii. There have been not many published paper reaching in chemical constituents of epimedium wushanense at present, especially, there have been not report the chemical constituents of roots of Epimedium wushanense T. S. Ying, and the root of it has been used to therapydisease as well as Coptidis phizoma in civilian.This paper have three major part:Part one1. The components were separated on Silica gel column, then repurified on Sephadex LH-20 to obtain 12 compounds. The structure was identified by spectral evidence of UV, IR, NMR, MS, COSY and NOE. The compounds, which are diphylloside A, epimedoside A, epimedin C, icariin, epimedoside C, icarisoside A, desmethylanhydroicaritin, oleanolic acid and F2, F3, F4 , F12 (they are appraiseing ). In addition to, diphylloside A, epimedoside A, epimedin C, epimedoside C, icarisoside A, desmethylanhydroicaritin, oleanolic acid were first obtained from roots of Epimedium wushanense T. S. Ying.2. By structure appraisement and HPLC comparement, identificated the structure of rouhuoside is mistake, it should be epimedin C.3. Established a HPLC method for simultaneous determination of the seven prenylflavones, diphylloside A, epimedoside A, epimediin C, icariin, epimedoside C, icarisoside A and desmethylanhydroicaritin in different organs (roots, stems, leaves), different area, different years of E. wushanense T. S. Ying, and each prenylflavone was also measured in leaves of ten Epimedii species (E. brevicornu Maxim., E. elongatum Komarov., E. mikinorii Steam., E. wushanense T. S. Ying, E. parvifollium s.z.et T. L. Zhang, E. pimedium sagittatum (Sieb. et Zucc.) Maxim., E. pubescens Maxim., E. ilicifolium Franch., E. dolichostemon Steam., E.koreanum Nakai.) from different culture areas. And compare the content variety of epimediin C and icariin in different growing seasons.We found the content of epimediin C is was higher than icariin in each organ (roots, stems, leaves) of Epimedium wushanense; the content of epimediin C is much higher than that of icariin in each month of the growing seasons of Epimedium wushanense, and epimediin C is the main component in Epimedium wushanense.4. We are successfully complete the task of People's Republic of China Pharmacopoeia of 2010.Part twoStudied the extraction process of barrenwortflavonoid from Epimedium wushanense with LSA-20 macroporousresin, The flavonoid extraction content of icariin was > 66.5%, and the extraction process is simple and convenient. The treatment of regenarated resin is easy, this method is advisable.Part threeWe searched the biological activities of seven prenylflavones, diphylloside A, epimedoside A, epimediin C, icariin, epimedoside C, icarisoside A and desmeth-ylanhydroicaritin.1. By study of antioxidant activity through Scavenging effects to DPPH radical, seven compounds have significant antioxidant activity. Desmethylanhydroicaritin and epimedoside C have shown higher antioxidant activity than the other five flavonoids. Scavenging effects of the two flavonoids were high as well as Vitamin C.2. By study of antibacterial experiment, the diphylloside A, icarisoside A and desmethylanhydroicaritin have significant activity for Pseudomonas. Ueruginosa. Icarisoside A and desmethylanhydroicaritin have significent antibacterial activity in solid nutrient medium, especially, diphylloside A has significent antibacterial activity in fluid nutrient medium for Pseudomonas. Ueruginosa (PAO ), this result is coincidence with its use as Coptidis phizoma in civilian. Thus, diphylloside A is better than ordinary antibiotic as a hoping antibiotic.
