Objective: to examine the effects of intra coronary injection of recombinant human basic fibroblast growth factor(rhbFGF)on treatment of acut myocardial infarction in swine models. Methods: The human bFGF pcDNA was cloned into enkaryontic expression vector pcDNA3. 15swines model was reproduced by opening chest and ligating the left circumflex(LCX)coronary artery. After model reproducing, rhbFGF was injected into coronary of the rhbFGF group to observe the effects of angiogenesis by pathology histological sections, angiography of coronary artery, and also calculated the left ventricular ejection fraction(LVEF) and left ventricular diastole and pressure (LVDEP), then estimate the control and gene therapy groups which were raise 2 weeks. Results: Changes of electrocardiogram testify that the model of AMI is successful. Histologic analysis found: there was a significant increase in the density of capillaries and arterioles in two gene therapy groups. After 4 weeks of occlusion, LVEF and LVEDP was significantly recovered from the derpressed value at the time after occlusion. Conclusion: introcoronary injection of rhbFGF gene canpromote angiogenesis and heart function in ischemic myocardium.
