Objective To study the feasibility of earlier pedicle division of random flap in the rats with local administration of adenovirus-mediated basic fibroblast growth factor (bFGF). Method The animals were 30 male Sprague-Dawley rats which were randomly divided into three groups.Group I ten rats were received replication-deficient recombinant adenovirus (Ad-bFGF), group II ten rats were received replication-deficient recombinant adenovirus carring Green fluorescein protein (Ad-GFP), group III ten rats were received Phosphate buffered saline (PBS). 2cm X 6cm dorsal random skin flap was designed with pedicle at the level of the iliac crest. The reagents were injected into the flaps during elevation.The pedicles were divided 4 days later. On the 7th day after pedicle division,all skin flaps were investigated the survival rate of the flap and skin samples from the distal survival portion of the flaps were collected for morphological and immunohistochemical analysis to detect the expression of bFGF and microvascular density. Results The flap survival rates of the groups I , II and III were (97.70±2.47)%,( 78.32±2.22)% and (79.21±2.29)% and density of microvascular sections was (27.32 ±4.30) / HP, (17.56 ± 3.08) / HP, and(18.48 ± 3.03) / HP. There was very significant difference between group I and the later groups(P<0.01). But there was no significant difference between group II and group III (P>0.05 ). Immunohistochemical staining documented increased deposition of bFGF in group I compared to the later groups. Conclusion The Ad-bFGF gene therapy is able to improve earlier pedicle division of random flap.
