Background and Objective At the beginning of 20th century,withthe invention of technique of surgical sewing method,especially theimmunosuppressant was used to inhibit immnunologic rejection.Thetransplantation has became probably in practice.In recentyears,allotransplantation is a wildly accepted theory for the treatment ofmany serious diseases.Because the supply of allografts is limited.It is hardto meet the increasing clinical demands.Due to the quanlity of some animalnumerous,growing cycle to be short It is easy to feed.Physiology andanatomy is similar to mankind.So many researcher utilize the animal organor tissue to perform xenotransplantation.But the immunolgic rejection isstill the grime problem we are faced with.It is the key point that how toavoid the immunologic rejection effectively at present. In the recent years,it is reported abroad that the valved bovine jugularvein conduit(VBJVC) tanned with glutaraldehyde be a promisingbiomaterial for the repair of right ventricale outflow tract.And there aresome study for the morphologic and hydrodynamic characteristics ofVBJVC in domestic,but has not been used in clinic yet.The research isaimed at to observe the antigenicity of glutaraldehyde VBJVC,in order tooffer the theoretical foundation for preparation of VBJVC and clinicalpractice. Methods 15 New Zealand white rabbits were randomly divided intothree groups,5 for each group.Group A:The glutaraldehyde tanned VBJVCtissue mixture suspension was injected into the rabbit's abdominal cavity.The glutaraldehyde tanned VBJVC tissue patches were buried in therabbit's back skin. Group B:Fresh VBJVC tissue mixture suspension wasinjected into the rabbit's abdominal cavity.The Fresh VBJVC tissue patcheswere buried in the rabbit's back skin. GroupC:Inject with the rabbit'sabdominal cavity and back skin of saline.Peripheral venous blood weretaken one day before the experiment,and at day 1,3,9,12,18,26 after theexperiment in all animals.IgG and IgM were measured by ratenephelomepry.Serum tumor necrosis factor(TNF)-α,interleukin(IL)-6,IL-8were measured by enzyme linked immunosorbent assays(ELISA),BuriedVBJVC tissue patches was obtained one month later in all groups formicroscope pathological and immunohistochemistry examination.Forstatistical analysis,repeated-measures analysis of variance was used formultiple brtween-group comparisons.Results are expressed with F,P.Pvalues<0.01 were considered significant. statistical analysis was performedusing SAS 8.02. Results There was no difference in measured results before theexperiment between the three groups.The expression of serum IgG,IgM andTNF-α, IL-6, IL-8 began to rise significantly in group B twelve days afterthe experiment.There were no obvious rose in measured results in group Aafter the experiment.There was no change in group C before and after theexperiment There were significantly difference in IgG,IgM and TNF-α,IL-6, IL-8 compared between group B with group A and group C.Noobvious difference was found between group A and group C.Buried tissuepatches histologic examination revealed that in group B there were obviousinflammatory response compared with the other groups.Immunohistochemistry examination:GroupB has more IgG,IgM,C3 todeposit,but group A and C seldem or almost have no IgG,IgM,C3 todeposit. Conclusion As one kind of raw material of xenograft,fresh VBJVChave obvious antigenicity.The glutaraldehyde tanned VBJVC significantlydecreases the antigenicity.Thus avoid the development of immunologicrejection.
