Objective To evaluate the relationship between single nucleotide polymorphism (SNPs) ±45T→G in adiponectin gene exon 2 and type 2 diabetes mellitus (T2DM), obesity or insulin resistance (IR) in Zhuang and Han people in Guangxi.Methods 616 subjects from the diabetic census population in Guangxi were divided into Zhuang and Han people groups by random stratified sampling. According to oral glucose tolerance test (OGTT), each group was divided into 3 subgroups: Zhuang-NGT (normal glucose tolerance, n=98), Zhuang-IGT (impaired glucose tolerance, n=108), Zhuang-T2DM (n=97) and Han-NGT (n=95), Han-IGT (n=103) and Han-T2DM (n=115). The T-G polymorphism in adiponectin (exon 2) was measured by polymerase chain reaction -restriction fragment length polymorphism (PCR-RFLP). Fasting insulin (FINS) were measured by radioimmunoassay. At the same time, BMI, waist circumference, hip circumference, TC, TG, HDL, LDL, UA, FPG, PPG and other related parameters were performed in all subjects. Insulin resistance index (IRI) was calculated in homeostasis model assessment (HOMA).Results ① The BMI, FPG, PPG, FINS, HOMA-IR and ISI levels in GG, TG and TT geotypes at position 45 in adiponectin (exon 2) in Zhuang subjects were respectively followed: BMI: 24.99 ± 3.19, 24.17±2.52, 23.73 ± 2.66 (kg/m~2), FPG: 7.99 ± 4.23,6.04 ± 2.42,5.97 ± 3.06 (mmol/l), GG genotypes were higher BMI and FPG levels than TT genotype (p<0.05, respectively). PPG: 11.24 ± 5.19,9.48 ± 4.29,9.21 ± 3.96 (mmol/1), FINS: 30.02 ± 47.96, 17.91 ± 39.47, 17.35 ± 21.93 (mU/l), HOMA-IR: 12.06 ± 22.77, 4.59 ± 9.09, 5.14 ± 8.46, GG genotypes were higher PPG, FINS and HOMA-IR levels than TG and TT genotypes(all p<0.05). ISI: 0.29 ± 0.03, 0.32±0.03, 0.33 ± 0.04, TT genotype was higher than GG genotype(p<0. 05). The HDL was significantly higher in TT genotype at position 45 in adiponectin (exon 2) than that in GG genotype in Han subjects( 1.44 ±0.48 nmol/1 vs 1.20 + 0.45 mmol/1, p<0. 05). ?The GG genotype in T2DM group was significantly higher than that in NGT and IGT groups (p<0. 05) in Zhuang subjects. Allele frequencies were no significant difference among three groups (p>0. 05). When adjusted for multiple factors (age, gender, BMI, WHR, FPG, PPG and FINS) of the SNPs+45GG genotype with reference to the TT genotype, the multiple adjusted OR was 3. 077(95%CI: 1. 240-7. 634) in T2DM group in Zhuang people. The multiple adjusted OR of the GG genotype with reference to the TT genotype was 3. 685(95%CI: 1. 277-11. 885) when IGT group and T2DM group merged into hyperglycemia group in Zhuang people.The gene frequencies were no significant difference among three groups (p>0. 05) at position 45 in Han subjects. Allele frequencies were no significant difference among three groups (p>0.05) in Han subjects. (3)The gene frequencies were statistical significance between non-obese~T2DM group and obese-T2DM group (p<0. 05) at position 45 in adiponectin (exon 2) in Zhuang subjects. The GG gene frequency was higher in obese-T2DM group than in non-obese~T2DM group. The gene frequencies were no significant difference among non-obesity subjects and obesity subjects in NGT, IGT and T2DM groups (p>0. 05) at position 45 in Han subjects. ?Adjusted for age and gender of the GG genotype with reference to the TT genotype, the OR of IR was 7.476(95%CI: 2.441-22.896) in Zhuang subjects and 2.876 (95%CI: 1. 141-7. 248) in Han subjects.Conclusions CD The SNPs +45 GG genotype in adiponectin gene has significantly higher BMI level than TT genotype in Zhuang people, which indicates GG genotype is related to obesity in Zhuang people in Guangxi. (2)The SNPs +45 GG genotypes in adiponectin gene have significantly higher FPG and PPG levels than TT genotype in Zhuang people, and GG genotype has significantly higher in T2DM than that in NGT and IGT groups, which indicates GG genotype is related to hyperglycemia and...
