Objective To study the expression of apoptotic cells and apoptosis-regulatory proteins Bax,Bcl-2, apoptosis induced factors and cytochrome C in renal cells of type 2 diabetic KKAY mice. Methods The mice were divided into two groups according to the levels of blood glucose: control group and diabetic group, with seven mice in each group. The mice in diabetes group were given high fat diet, while the mice in control group were fed with a standard diet with full nutrition. The mice were sacrificed at the third month. The numbers of apoptotic renal cells were detected using terminal deoxynucleotidyl transferase (TdT)-mediated dUTP-biotin nick end labeling (TUNEL)techniques and ultrastructure were observed by electron microscop . The expression of apoptosis-regulatory proteins, Bax ,Bcl-2,AIF and CytC were assayed using immunohistochemical techniques. Results Apopotic cells in diabetic mice were significantly greater than those in control group (P<0.01). Compared with control group, protein Bax, AIF and Cyt C were observed to increase in diabetic mice (P<0.01), while Bcl-2 and BCI7 BAX decreased (P<0.01).Conclusion Apoptotic cell numbers increased in diabetic KKAY mice. the multiple factors induce apoptosis in kidney of diabetes, which mediate, at least in part, by altering the BCL family and releasing AIF and cyt c .in other words ,mitochondria play a role in the regulation of apoptosis in kidney of diabetes.
