Objective: To study the behavior of chondrocytes cultured in biological fibrin glue and in biological fibrin glue/cancellous bone matrix in vitro and to investigate the effect of the two scaffolds engineered with chondrocytes transplantation for the repair of articular cartilage defects. Methods: (1) Chondrocytes taken from New Zealand rabbits aged 3 weeks were cultured in biological fibrin glue and biological fibrin glue/cancellous bone matrix in vitro. (2) Phase contract microscopy, electron microscopy and histology were used to study the behavior of chondrocytes cultured in biological fibrin glue and biological fibrin glue / cancellous bone matrix. (3) Repair full-thickness articular cartilage defects of rabbit by chondrocytes/biological fibrin glue or chondrocytes/biological fibrin glue/cancellous bone matrix. The defects were only implanted with biological fibrin glue/cancellous bone matrix or left empty in control groups. The rabbits were killed in 1,2 and 3 months after operation for gross, histological and TEM examinaton. Results: (1) Most chondrocytes retained their oval shape when cultured in biological fibrin glue or biological fibrin glue/cancellous bone matrix, a few on the surface of the two scaffolds appeared as they did in monolayer culture. (2) The number of chondrocytes in the two ascaffolds both increased significantly after 9 days; The survival rate was 95%. (3) AB-PAS stain (+), Masson's stain (+).(4) Biological fibrin glue was dagrade within 1 months after operation and Cancellous bone matrix was dagrade within 2 months after operation. Cartilage defects were repaired by cartilage tissue, articular cartilage and subchondral bone were soundly healed. Chondrocytes and cartilage matrix in repaired tissue were found to be resembled the normal. The defects implanted with biological fibrin glue/cancellous bone matrix and the empty group was repaired by fibrous tissue. Conclusion: Chondrocytes grow well and can synthesize cartilage extracellular matrix in biological fibrin glue and biological fibrin glue/cancellous bone matrix, Articular cartilage defects could be repaired by the two scaffolds engineered chondrocytes,...
