Objective: To detect the expression and study the role of survivin , EphB4 and p-stat3 in hamster buccal pouch carcinogenesis. Methods: 96 hamsters were divided into four groups, including one normal group and three experimental groups, there were 12 hamsters in each group. All the hamster buccal pouch carcinogenesis was induced by 0.5% DMBA.The three experimental groups were killed after 3 weeks,9 weeks and 12 weeks separately,4 hamsters were killed each time. We obtained 90 samples except 6 insignificant samples, including 23 cases of normal oral mucous,22 cases of hyperplasia epithelia,20 cases of dysplasia epithelia and 25 cases of oral squamous cell carcinoma(OSCC). The expression of survivin and EphB4 was detected by RT-PCR,the expression of p-stat3 and survivin protein was detected by immunohistochemical assay in the tissues above. Results: The positive expression of survivin mRNA was in an increasing gradient from normal oral mucosa group, epithelia hyperplasia group, epithelia dysplasia group to OSCC,the positive rate was 0,13.6%,45.0% and 76.0% in each group above. There was no significant difference in survivin expression between normal epithelia and hyperplasia epithelia(P>0.05),but a statistically significant difference was found between any two groups of the rest(P<0.05). The positive expression of EphB4 mRNA in normal oral mucosa group, epithelia hyperplasia group, epithelia dysplasia group and OSCC was 87.0%,90.9%,85.0% and 96.0%. There were no significant...
