A high performance liquid chromatography (HPLC) analysis method for determination of diester-diterpenoid aconitum alkaloids from aconite roots after processing, heating and combination was developed. The analysis was carried out by HPLC with a C18 column (250 mm×4.6 mm,5 μm) which was connected with an analytical guard column (7.5 mm×4.6 mm,5 μm), methnol-0.3%triethylamine was used as mobile phase and eluent was detected at 235nm. The column temperature was 40 ℃and the flow rate was 0.8 ml/min. The calibration curves were linear in the range of 5.65~113 μg/mL for mesaconitine(y=18795.5x-36075, r =0.9997), 6.5~130 μg/mL for aconitine(y=17485.4x-13386.7, r =0.9993) and 5~100 μg/mL for hypaconitine(y=20287.9x-28543.4, r=0.99994), respectively. This method showed good recovery, high sensitivity, satisfactory accuracy, specificity and stability. The content value of diester-type alkaloids in the different samples is unprocessed Caowu > unprocessed Fuzi> Zhicaowu> Heifupian> Danfuzi> Baifupian> Paofuzi. The content of diester-type alkaloids in unprocessed Caowu among them was highest and the content of alkaloids in Zhicaowu among the processed aconitum herbs was highest. The processing conditions were optimized by orthogonal tests. For hypaconitine, the optimal conditions of processing by steaming was to dip for 5 h, to steam for 8 h, and then to dry 16 h at 80 ℃while the optimal conditions of processing for mesaconitine was to dip for 9 hours, to steam...
