Objective Study on the effects of diesterifiable alkaloids and monoester alkaloids in Radix Aconiti processed by different conditions. To observe and explore the variation trend and rule of diesterifiable alkaloids and monoester alkaloids in different high temperature steaming system, and obtain optimal high-temperature steaming process for the different quality of Radix Aconiti.Methods The content of diesterifiable alkaloids and monoester alkaloids was measured by HPLC method. We observed the changes of two kinds of alkaloids when the same steaming process method applied, and analyzed the variation trend of diesterifiable alkaloids and monoester alkaloids in different high temperature steaming system. And then we choosed the optimal high-temperature steaming process for the different quality of Radix Aconiti with investigation factors of time, temperature of steaming and softening method.Results clarify initially the correlation between quality and processing conditions of Radix Aconiti. The content of diesterifiable alkaloids decreased, but the content of monoester alkaloids increased firstly and then decreased when processed by high temperature steaming. The best technology for high-temperature steaming is:steaming was applied in 120℃ with Vacuum run for half of an hour when the content of diesterifiable alkaloids is 0.05%-0.09%; steaming was applied in 100℃ with Vacuum run for one hour when the content of diesterifiable alkaloids is 0.09%-0.13% and steaming was applied in 120℃ with Vacuum wetting for 1.5 hours when the content of diesterifiable alkaloids is 0.13%-0.17%. The content of monoester alkaloids and the diesterifiable alkaloids in the samples processed by different high temperature steaming conditions.are both qualified for the Chinese Pharmacopoeia edited in 2010. The results of amplification experiments are qualified for the requirements of industrial production on the basis of the above research work.Conclusions This experiment confirmed the correlation between quality and processing conditions of Radix Aconiti. Refine the levels of diesterifiable alkaloids and choose the optimal high-temperature steaming process by orthogonal design. The more reasonable and time-saving processing technology was obtained. We also provide a new method of Processing technology for Aconitum.
