Object To investigate the changes of concentration of NO, SOD and Ca ~2+ in the serum, the expression of the apoptotic gene Bcl-2, Bax and caspase-3 in the small intestine and the remote organ of the lung and heart. Try to find out the apoptotic mechanism of small intestine, the remote organ of the lung and heart after small intestine ischemia reperfusion.Methods Small intestine ischemia reperfusion injury models were established in 48 Wistar rats. According to different ischemic and reperfusive time, 8 groups(N=6/group) were divided, including normal control, ischemia time 40 min, reperfusive time 30, 60, 120 min, 1, 3, 7d groups. The portal blood were taken to exame the concentration of Nitric Oxide (NO), Superoxide Dismutase(SOD), Calcium on different pionts. Simultaneously, the expression of the apoptotic gene Bcl-2, Bax, Caspase-3 in the small intestine and the remote organ of the lung and heart was observed by the irnmunhistochemical SP or SABC method.Results There were increasing of the level of NO in the serum from ischemia time 40 min to reperfusive time 1d group, meanwhile, the concertration of SOD was decreasing (P<0.05). The level of NO in the serum began to decrease gradually on 3 and 7d, meanwhile, the concertration of SOD was increasing. The chang of the concertration of Calcium in the serum was not significant among 8 groups(P>0.05). The ratio of positive cells of caspase-3 in the small intestine, the lung and heart began to up-regulate with time. The top point was on reperfusive time120 min and 1d group. The ratio of positive cells of Bax began to up-regulate with time also. The peak point was on reperfusive time 1d, 30 min and 120 min group. The ratio of positive cells of Bcl-2 in the small intestine, the lung and heart began to down-regulate with time. The lowest point was on reperfusive time 60min and 120 min group.
