Study On The Biotransformation Conditions And Effects Of Yupingfeng Powder

Objective: In order to find new activity or improve the effective of Yupingfeng Powder ( YPFP) , it was transformed by Saccharomyces cerevisiae. And this experiment mainly focused on the biotransformation conditions. Methods: We selected two strains of Saccharomyces cerevisiae Siand S₂ to transform YPFP and compared the effects of the transformed products. At the same time, we studied the biotransformation time. The efficacy of the transformed products were tested by assaying the chicken erythrocy- te phagocytosis rate and index of peritoneal macrophage(PMφ) after mice were given the transformed products. Since microorganism can have different biotransformation activities in different mediums, we studied the relationship between the medium and the biotransformation system. Three different culture mediums, PDA, YPD and malt, were studied. Then the effects of the transformed products were compared by the means mention above. And we further studied the effects of the transformed products on the proliferation activity of PMφ, T and B lymphocyte of spleen in mice in vitro. By using the methods of serum pharmacology, the drug-containing serum was extracted, Serum contained transformed or untransformed YPFP were given to those cells and then MTT was added separately. The absorption values (A) were assayed by a enzyme-labeled machine. We also studied the anti-tumour effect of the transformed products on a mice model loaded H₂₂ cells in vivo. And the anti-tumour index was assayed by weighing the weight of the tumour. Results: Several conditions of biotransformation have been established.
