The Mechanism Of Treating HSK Mice Model With Yupingfeng Powder

Purpose: To investigate the mechanism of Yupingfeng Powder in curing mice with infection of herpes simple virus type 1 (HSV-1).Methods: Sixty Balb/c mice were divided into three groups randomly: normal control group, treatment group and model group. The herpetic simplex keratitis (HSK) mice model is founded through corneal scarification infected with HSV-1. In treatment group, Yupingfeng Powder apozem were used by intragastric administration; In model group , physiological saline were used by intragastric administration; In normal control group, the mice are left to eat and drink by themselves in the same environment. Observe and analyze the incidence, death-rate and clinical features of the three groups on the 1st, 3rd, 5th, 7th, 14th days respectively after topical HSV-1 corneal infection. 14days after HSV-1 infection, the expressions of interleukin-2(IL-2), interleukin-4(IL-4) and interferon gamma(IFN-y) in mice blood-serum were detected by the method of ELISA.Results: The death-rate of treatment group was 15%, model group 16.67%. the statistically slight difference between these two groups indicates that Yupingfeng Powder exerts no influence on the death-rate in HSK.The results of ELISA reveals that the IL-4 level in mice blood-serum of the model group is noticeably increased (P<0.01) in contrast with that of the normal control group; the IL-2 level and IFN-γlevel are noticeably decreased in contrast with that of the normal control group. While the Yupingfeng Powder treatment group, after the treatment, the level of IL-4 (P<0.01) in mice blood-serum is conspicuously lower than that of the model group, and its IL-2 level and IFN-γlevel (P<0.01) are noticeably increased.Conclusions: After HSV-1 corneal infection, the level of IL-4 in mice blood-serum would increase, but IL-2 and IFN-γwould decrease. After the treatment through Yupingfeng Powder apozem, the level of IL-4 could be lowered, and IL-2 and IFN-γcould be increased. The balance of Th1/Th2 was restored. Yupingfeng Powder can relieve the corneal lesion, accelerate the recovery and lessen the recurrence risk.
