Chaihu the root of bupleurum Chinense DC. , belonging to the family of Umbelliferae, is the high qualified herbal drug grown in northeast part of China. It has the relieving exterior syndromes by diaphoriesis, soothing the depressed liver and improving the functional aspect of internal organs.In this paper, the method for evaluating the quality of cultivated Chaihu has been established by using RP-HPLC and spectrometry, saikosaponin-a, c, d and total saikosaponins were taken as the marker substance. By the method, the quality evaluation of various kinds of Chaihu, generated from different cultivating area, collecting time and processing method, has been carried out.As result, Chaihu cultivated in Qitaihe county has a much better quality than the others, Qitaihe was considered as the proper areas for cultivating the herbal drug. Among the samples collected at different seasons, that collected during the first 10 days of April has a highest content of the four marker substance, the collecting time was set on the early spring of the third growing year. For the processing method, drying at shade is the best selection. The determined results of the samples cultivated in different fertilized measures showed that there was no difference in different fertilized measures. The determined results of the samples in different grade showed that the first grade sample is the best in quality. The determined results of the samples cultivated in different density showed that the contents of saikosaponin-a, c, d and saikosaponins were hightest in the sample cultivated in density of 50 plant/m~2.
