Essential oil and Saikosaponin are active constitutent of Bupleurum chinense, The traditional extraction of essential oil of Bupleurum only to be considered to extract the essential oil, but ignore the gruffs which had been taken essential oil can also been taken Saikosaponin, that will lead to waste of resources. We not only consider the extraction rate of essential oil, but also consider the extraction rate of Saikosaponin. We used both the extraction rate of essential oil and the extraction rate of Saikosaponin as the indicators to determine the best conditions of extraction of essential oil of Bupleurum. The conditions are as follows: dipping time (h),concentration of NaCl and extraction time (h). We found that C3 NaCl dipping for T1, then 100℃extraction for D3 is the best conditions, and the best conditions of extraction rate of Saikosaponin is 8 times 70% alcohol, 80℃back flow extraction for 3 times. So we can take the high extraction rate of essential oil and high extraction rate of Saikosaponin. This industrial Arts is more useful in practice.Essential oil and Saikosaponin has been known as the most effective components of Bupleurum chinense. We use both the Essential oil and Saikosaponin as compatibility will take joint action. Saikosaponin has high haemolyticus, so we can't make it as injections. And in the area of acidity, Saikosaponin isn't stabilize. It will decimpose in gastric acid. So Saikosaponin can't be absorbed in gastrointestinal tract. Essential oil of Saikosaponin is almost used as injections, but these years there were many adverse reactions appearance. In order to educe the activity of relieving fever,antiinflammatory and so on. We let Essential oil and Saikosaponin absorb in buccal cavity. And then Saikosaponin will be not decimpose in gastric acid, and it will avoid the first pass effect.The instability of essential oil of Bupleurum chinense is poor, we usedβ-CYD to embed it to increase its stability. And it will be absorbed quickly to educe the activity of relieving fever,antiinflammatory,and so on. We establish the methodology of UV- visible light spectrum to determine the inclusion rate of essential oil of Bupleurum chinense. Essential oil of Bupleurum chinense at 270nm has maximum absorption, and good linearity. We used yield and inclusion rate of inclution compound as the indicators, through single-factor and orthogonal experimental to determine the best conditions of magnetic stirring solubilization. The results are as follows: the best ratio of essential oil andβ-cyclodextrin is B3. Small amount of ethanol dissolve the volatile oil of Bupleurum chinense, and then slowly added to the A3β-cyclodextrin solution ,which is dissolved in high temperature, C3 constant temperature stirring D2, cool to room temperature, iced storage for 10h, till theβ-cyclodextrin totally seperate out, filtering, 60℃oven-drying.Saikosaponin has poor dissolubility in water. We use M to increase it's dissolubility in water. We establish the methodology of HPLC to determine the solubility of Saikosaponin-a and Saikosaponin-d. Methodological conditions are as follows: The stationary phase: Diamondsil C18 (250 * 4.6mm, 5μm); The mobile phase: acetonitrile - water (38:62); flow rate: 1.00ml/min; column temperature: room temperature. We used solubility of Saikosaponin a and Saikosaponin d as the indicators, through single-factor and orthogonal experimental to determine the best conditions of magnetic stirring solubilization. The results are as follows: sample of Saikosaponin(purity> 50%): M(w/w) is A2, B2 times 70% ethanol than the sample of Saikosaponin(purity> 50%) dissolve sample, and then slowly adding the solution to 0.1g/ml M, C3 constant temperature stirring 1h, drying, grinding and screening. We found that the solubility of Saikosaponin a and saikosaponin d in water increase for 25 times and 52 times.
