The distribution of the Bupleurum plants is wide with many types. As the variety is disordered and people have different habits of drug usage, the quality of bupleurum and its preparations are not stable."Chinese Pharmacopoeia" only includes the dried root of Bupleurum chinense DC. and Bupleurum scorzonerifolium Willd. In the current drug standard of our country,549of the drugs preparations use radix bupleuri medicinal materials, and then these medicinal materials present demand over supply. Bupleurum marginatum Wall. ex DC. is one of the three kinds of Bupleurum medicinal materials with long history of pharmacy and abundant resource implications. As it is not recorded by "Chinese Pharmacopoeia", the application is limited, so that the resources cannot be effectively used. Therefore, this topic will take Bupleurum chinense DC. as the reference medicinal materials, establish quality evaluation system of bupleurum plants, and systematically analyze its chemical components, pharmacology and pharmacodynamic effects, make comparison between it and bupleurum marginatum to illuminate the possibility and reliability of being used as radix bupleuri medicinal materials. Around this objective, this topic makes the researches as below:1. Resource survey and identification of B. chinense DC.and B. marginatum Wall. ex B. chinense DC. and B. marginatum Wall. ex have prominent identification features which can be used to distinguish them, including the texture of root,"membrane-bounded sacs"of B. marginatum Wall. ex,total bracts pulses, length of leaves, quantity of pulse, quantity of total bracts pulses, and the comparison of the length of small bracts and pedicels.Medicinal materials can be distinguished from the root quality. It’s unable to distinguish between slices and powder.2. The assessment research on the volatile oil of bupleurum marginatum and saikosaponin.Using SPME-GC-MS technology to set up volatile oil detection method, B. marginatum Wall. ex with48ingredients are identified, B. chinense DC. with21ingredients are identified, B. malconense Shan et Y. Li with32ingredients are identified, B. microcephalum Diels. with32ingredients are identified, B. longicaule var. amplexicaule C.Y.Wu with15ingredients are identified. Besides, compare the similarities and differences of Volatile oil chemical composition between B. marginatum Wall. ex and B. chinense DC., get10common compounds, and prove that n-hexadecane acid and9,12-18octadecadienoic acid are major ingredients for the volatile oil of radix bupleuri.In addition, make similarity calculation and clustering analysis towards30samples.3. The assessment research on the Saponin compounds of B. chinense DC.and B. marginatum Wall. exUsing UPLC-Q-TOF technology to set up saponin compounds detection method, B. marginatum Wall. ex with28compounds are identified, B. chinense DC. with38compounds are identified, B. malconense Shan et Y. Li with32compounds are identified, B. microcephalum Diels. with31compounds are identified, B. longicaule var. amplexicaule C.Y.Wu with32compounds are identified. Also it compares the components differences between B. chinense DC.and B. marginatum Wall. Ex., and marks the16mutual components of the both. Besides, it conducts similarity calculation,assaying and cluster analysis to the64batch Bupleurum samples. The results show that the quality of B. marginatum Wall. ex. is steady and controllable while other Bupleurum medicinal materials from different resources also have improvable space under the premise of quality control.4. The assessment research on the pharmacological activity of B. chinense DC.and B. marginatum Wall. ex.The big mice fever model caused by2,4dinitrophenol, the big mice toes swelling model caused by carrageenin and the small mice auricle swelling model caused by xylene are employed to observe the antipyresis and anti-inflammatory effects of B. chinense DC.and B. marginatum Wall. ex.. The results show that the B. chinense DC.and B. marginatum Wall. ex. have an obvious inhibiting effect to big mice toes swelling and small mice auricle swelling,and it can reduce the fervescence effect caused by2,4dinitrophenol. All the different dosage groups of B. chinense DC.and B. marginatum Wall. ex. tend to have the antipyresis and anti-inflammatory effects. As to the antipyresis effect, there’s no dosage relativity between those two. The results tell us that bupleurum marginatum and saikosaponin both have a good antipyresis and anti-inflammatory effect,and B. marginatum Wall. ex. can be substituted with B. chinense DC. clinically.
