Objective:To study the expression levels of Midkine(MK),Ki-67 and CD34 in giant cell tumor of bone and their prognostic value.Methods:The expression levels of MK in 37 cases of giant cell tumor of bone were studied by S-P immunohistochemisty method,while expression levels of Ki-67 and CD34 were studied by Envision method.Their correlation with clinicopathological features and recurrence state were also investigated.Results:(1)Overexpression of MK were found in giant cell tumor of bone.Its expression ratio was 51.35% .The expression of MK were not correlated with its histograde , radiological grade and recurrence state(P>0.05).But MK positive cases had higher Ki-67 index and microvessel density than MK negative cases (Ki-67 index: 139.95 ±48.87 vs 98.29 ± 31.60; microvessel density: 192.57 ± 20.16 vs 145.53 ± 25.83,P<0.01).(2)The average Ki-67 index in giant cell tumor was 118.86 ± 46.16,pathological grade Ⅲ cases showed higher index than grade Ⅰ and grade Ⅱ cases(P<0.01),but there was no significant difference between grade Ⅰ and grade Ⅱ cases.When radiological grade was concerned, there wassignificant difference between grade III cases and the remaining two groups(P<0.05, PO.01,respectively). Recurrent cases had a higer Ki-67 index than cases without recurrence(161.33 ± 29.84 vs 93.80 ± 32.80,P<0.01). (3)MVD in giant cell tumor of bone was 169.44 + 32.92, there was significant difference between histological grade III and gradel cases(202.48 +14.45 vs 159.25 + 28.26 ,P<0.01),no significant difference was found between grade III and grade II cases. Recurrent cases had higer MVD than cases without recurrence(193.56 + 21.96 vsl54.94 + 29.82,P<0.01).Conclusion:Overexpression of midkine was found in giant cell tumor of bone.MK may invoveled in tumor cell proliferation and angiogenesis,but was not correlated with tumor recurrence. Correlation between MVD and tumor cell proliferation with tumor recurrence were suggested.
