Objective: Study the expression of CD146in giant cell tumormicrovessel.To investigate the correlation between microvessel density(MVD)marked with CD146, expression of vascular endothelial growth factor(VEGF)and the histological grade,radiological stage,recurrence state of the tumor,such as the clinical data.To intend to find possible prognostic indications ofthe aggressive behavior of the underlying pathology.Methods: Collect more than24months and follow-up records the thirdhospital of hebei medical university from January2008to January200839cases of giant cell tumor of the specimen that had line lesions curettage.Allcases were confirmed by pathological and imaging data, and Jaffeclassification and grading Companacci.They were20males and19femalesin this group. They were aged from18to69years with an average age of37.51.Tumors were located at Around the knee joint in20patients(distalfemurs in8,proximal tibia in12);other part in19.According to Campanacci’sradiological image classification,18cases classified grade I,16cases gradeII,5cases grade III.According to Jaffe’s histological grade,19cases classifiedgrade I,13cases grade II,7cases grade III.14case were recurrence and25cases were no.The expression of CD146and VEGF in39cases of giant celltumor of bone wase studied by using SP immunohisto-chemical method andanalysing MVD of CD146positive microvessel.Results:(1) The average MVD marked by CD146in giant cell tu-mor of bone was27.26±6.58. No significant difference was found among Jaffe grade while MVD marked by CD146rise step by step.According to Campanacci’s classification, stage3(aggressive type) was significanthigher MVD than stage1(quiescent type) and stage2(activity type),and that the difference was statistically significant(P<0.05).Recurrent casehad higher MVD(32.67±4.39) than cases without recurrent(24.22±5.60),an d that the difference w-as statistically significant(P<0.05).(2)VEGF express positive in most giant cell tumor of bone(26cases66.67%).No statistically significant was found among Jaffe grade(P>0.05). According to Campanacci’s classification,expression levels of VEGF was found rise stepby step,and that the difference was statistically significant(P<0.05).Recurrent case(92.86%) had higher positive rate of VEGF than cases without recurrent(52.0%),and that the difference was statistically significant(P<0.05).(3)VEGF positive cases(26cases) had higher MVD(29.18±6.47) thanVEGF negative cases(23.40±5.07),and that the difference was statisticallysignificant(P<0.05).There was a positive correlation(r=0.420P=0.008) between MVD and VEGF.Conclusion: CD146and VEGF are highly expressed in the giant celltumors of bone tissue, and is closely related to tumor angiogenesis.Both werepositively correlated, prompt CD146and VEGF in giant cell tumorangiogenesis in the process of new promote each other.VEGF expression andMVD marked by CD146were correlated to Campanacci’s classification andrecurrence,not to Jaffe grade,age,sex,tumor size and pathogenic site. So MVDmarked by CD146and VEGF can be used as a giant cell tumor of boneaggressive ability and prognosis of objective indicators.
