Objective To obtain the allele and haplotype frequencies of the three novel Y-STR loci (DYS508, DYS516 and DYS520) and discuss the possibility of applying them to forensic science by investigating these 3 Y-STR loci in Chengdu Han ethnic group.Methods 3 novel Y-STR loci were retrieved from the GDB database and amplified with PCR. The products of PCR were analyzed with polyacrylamide gel electrophoresis. Each allele of these 3 Y-STR loci was sequenced and the allele ladders were constructed. Alleles of these Y-STR loci were nominated according to recommendations of the International Society of Forensic Genetics. Results In our population studied, the gene diversity of DYS508, DYS516 and DYS520 was 0.7242, 0.7931 and 0.7374 respectively. The diversity of haplotype for these Y-STR loci was 0.9851. The value of discrimination power and the exclusion chance of paternity for these Y-STR loci were same as the value of haplotype diversity.Conclusion All of the 3 Y-STR loci are genetic polymoephic. The haplotype diversity shows good discrimination power and high exclusion chance of paternity. They are benificial to the test of male-female mixtures and paternity identification.
