Objective We have investigated a novel Y-STR locus(DYS714)on male Y chromosome to obtain the allele frequencies of DYS714 in Han population,Shanxi Province,P.R.China and to evaluate its application of forensic casework.Methods The candidate of Y-STR loci were selected from the GDB database.Primer pairs were designed on both side of the repeat region.The PCR products were analyzed with polyacrylamide gel electrophoresis.Each allele of this Y-STR loci was sequenced and the allele ladders were constructed.Alleles of these Y-STR loci were nominated according to recommendations of the International Society of Forensic Genetics.Results Our results show that the DYS714 locus consists of repeats of a core sequence (TTTTC).Five alleles were found among 120 males in Shanxi Province,The alleles are named 10,11,12,13,14and the allele frequency of 0.15,0.1583,0.2167,0.3333,0.1416 respectively. Genetic diversity(h)of DYS714 was 0.7808.The power of discrimination(DP)and the power of exclusion(PE)were both 0.7808.No amplified products could be detected from female sample.The typing results of different tissues from one individual were identical.Mutations were not found from 15 father-offspring pairs.Conclusion DYS714 have a high level of genetic polymorphism.This polymorphic system is very useful in forensic casework and human genetics research.
