Objective We have investigated a novel Y-STR locus (DYS718) on male Y chromosome to obtain the allele frequencies of DYS718 in Han population, Shanxi Province, P.R.China and to evaluate its application of forensic casework.Methods EDTA-blood specimens were collected from 116 unrelated males in Han population in Shanxi. The blood of 20 females were also collected as control.The genomic DNA of blood, muscle, heart, liver and kidney from a corpse were analyzed. Fifteen father-offspring pairs were investigated. Different male animals and different tissues (including blood, muscle, et al) of one corpse were analyzed. All blood-specimens were anticoagulated with EDTA ,the Genomic DNA was extracted via method of phenol-chloroform and the alleles of DYS718 were amplified by PCR. The PCR products were analyzed on 8% non-denaturing polyacrylamide gel electrophoresis followed by silver-staining. Each allele of the DYS718 was sequenced and the allele ladders were constructed. Alleles of the Y STR loci were nominated according to recommendations of the International Society of Forensic Genetics. The core repeats and their repeating numbers of these alleles were confirmed by sequencing.Results Our results show that the DYS718 locus consists of repeats of a core sequence (TTA). Five alleles were found among 116 males in Shanxi Province, The alleles are named based on the number of repeats. The allele frequency of 14,15,16,17 are 0.1207,0.3621,0.4138,0.1043 respectively. So calculated Genetic diversity (h) of DYS718 was 0.6782. The power of discrimination (DP) and the probability of exclusion (PE) were both 0.6782. No amplified products could be detected from female sample. The typing results of different tissues from one individual were identical. Mutations were not found from 15 father-offspring pairs.Conclusion It shows that DYS718 in Shanxi Han population is high valuable in the application of forensic casework and population genetics .
