| [Backgroud and purpose]:Bronchial asthma has often been proned to myocardial injury from the perspective of pathophysiology. The author once studied 12 cases of sudden death because of bronical asthma and found myocardial atropsy, necrosis and cicatrisation besides pulmonary injury. The mechanism of myocardial lesion of asthamtic patients is still unknown. In addition, some scholars report that the common drug - adrenaline can induce apoptasis of myocardium. To find the mechanism of myocardial lesion, the asthmatic model of rats was built on the basis of which different drugs were used to release the syndrom of asthmatic hypoxia.[Methods]:One hundred and twenty male Sparague-Dawley rats were randomly divided into normal control, experimental control, a group of no drugs to deal with asthmatic syndrom, a group of adrenaline to deal with asthmatic syndrom, a group of amoniphylline to deal with asthmatic syndrom, a group of adrenaline and amoniphylline to deal with asthmatic syndrom. Every group was divided as four subgroups according to experimental time (2w, 4w, 6w, 8w). Every subgroup hadfive rats. Then the rats were killed according to the time elapsed 2w, 4w, 6wor 8w after experiencing different treatment. The expression of p53 and caspase-8 were labeled by immunohistochemistry and the results were analysed in IMAGE analytical system. [Results]:(1) p53 and caspase-8' s expression of the male SD rats' myocardium of the common and experimental control group was negative. (2) Area and the multiplication of area and dentisity of positive signal of myocardium of all experimental groups was statistically higher than that of the common control and experimental control groups ( p<0. 01) on week 2. the trend of the expression of p53 and caspase-8 continuously increased with the time elapsed , till the 8th week, the area and the multiplication of area and dentisity reached the apex of this research. (3) the expression of p53 and caspase-8 of the group of adrenaline to deal with was statistically higher than other experimental groups (p<0. 05) ;the expression of p53 and caspase-8 of the group of adrenaline and amoniophylline to deal with was the least among the experimental groups(p<0. 01). the expression of the group of no drugs to deal with was to some extent higher than the group of amoniophylline to deal with(p<0.05). [Conclusion]: 1. The asthma itself had conduced to myocardial lesion.2 Adrenaline aggravated the degree of myocardial lesion of asthmatic patient, but amoniphilline has no bad effect on myocardium.3 The thearapeutic efficacy of the group of adrenaline and amoniphilline was good.Moreover, the myocardial lesion can belightened. Using small dose and many different drugs combined toutilize could both release the asthamatic syndrom and reduce thecomplications of drugs.4 The detection of p53 and caspase-8 could offer the auxiliarydiagnosis of sudden death of patients with bronical asthma in thework of forensic medicine. |
