Objective: To discuss in vitro inhibition effect of Laggera pterodonta Pomegranate Rind Taraxacum mongolicum on H.pylori metronidazole resistant strains and susceptible strains; Filter the best combination of anti-Hpylori ; Observe the effects of the combination of three drugs to H.pylori infected BALB/c mouse.Methods: (l)The Agar Dilution Method detect the minimal inhibitory concentration(MIC) of the water extract of Laggera pterodonta the water extract of Pomegranate Rind the water extract of Taraxacum mongolicum on H.pylori metronidazole resistant strains and susceptible strains, MIC50 MIC75 MIC90 were also calculated respectively. (2) Orthogonal Test Method filter the best combination of Natural Chinese drugs of anti-H.pylori metronidazole resistant strains and susceptible strains;(3) To establish a infection model of BALB/c mouse with Yunnan strains: l-139(H.pylori metronidazole resistant strains) and 1-1A5(H.pylori metronidazole susceptible strains); (4)To investigate the effects of the best combination of nature Chinese drugs in the treatment of H.pylori infected BALB/c mouse.Results: (1) MIC of Laggera pterodonta on H.pylori was in a range of 0.003125g/ml to 0.2g/ml, The susceptibility of H.pylori metronidazole resistant strains and susceptible strains on Laggera pterodonta has no statistical difference (relative median potential = 1.1338, 95% credible range from 0.60831 to 2.41187), MIC50 was 0.045g/ml and 0.043g/ml respectively, MIC75 was 0.117g/ml and0.091 g/ml respectively, MIC90 was 0.278g/ml #1 0.179g/ml respectively.MIC of Pomegranate Rind on H.pylori was in a range of 0.00390625g/ml to 0.5g/ml, The susceptibility of H.pylori metronidazole resistant strains and susceptible strains on Pomegranate Rind has no statistical difference (relative median potential^ 1.0275, 95% credible range from 0.38659 to 3.01571), MIC50 was 0.099g/ml and 0.095g/ml respectively, MIC75 was 0.327g/ml and 0.348g/ml respectively, MIC90 was 0.960g/ml and 1.116g/ml respectively.MIC of Taraxacum mongolicum on H.pylori was in a range of 0.00625g/ml to 0.8g/ml, The susceptibility of H.pylori metronidazole resistant strains and susceptible strains on Taraxacum mongolicum has no statistical difference (relative median potential 1.0890, 95% credible range from 0.53850 to 2.45446), MIC50 was 0.137g/ml and 0.136g/ml respectively, MIC75 was 0.426g/ml and 0.460g/ml respectively, MIC90 was 1.184g/ml and 1.372g/ml respectively.(2) The best combination scheme of the effect of the tree Nature Chinese drugs on the H.pylori metronidazole susceptible strains was : Laggera pterodonta(0.278g/ml): Pomegranate Rind(0.960g/ml) .-Taraxacum mongolicum(0.137g/ml); The best combination scheme of the effect of the tree Nature Chinese drugs on the H.pylori metronidazole resistant strains was: Laggera pterodonta(0.179g/ml): Pomegranate Rind(l. 116g/ml): Taraxacum mongolicum(0.460g/ml).(3) The infection model of BALB/c mouse with H.pylori strains 1-139 and 4-145 was successfully established by surgical method.The infectious rate is 90%(9/10) and 80%(8/l 0) in infection model group with the resistant strains and in one with susceptible strains respectively after 4 weeks, so the infectious rate is 85%(17/20); The infectious rate is 100%(10/10) and 100%(10/10) in infection model group with the resistant strains and in one with susceptiblestrains respectively after 8 weeks, so the the infectious rate is 100%(20/20); (4) 4 weeks after the treatment with the best combination of Nature Chinese drugs, in the metronidazole resistant group: The eradicative rates of Nature Chinese drugs (Laggerapterodonta+Pomegranate Rind+Taraxacum mongolicum ) > Nature Chinese drugs +double therapy(Colloidal bismuth subcitrate + tetracycline haydrochloride ) and triple therapy (Colloidal bismuth subcitrate+ metronidazole+tetracycline haydrochloride)on H.pylori is 60% > 86.7% and 40%.Nature Chinese drugs+double therapy is better than triple therapy,there is a statistical difference in the eradicative rates on H.pylori between then\(P<0. 05);There is no statistical difference in the eradicative rates between Nature Chinese drugs and triple therapy(P>0. 05); There is no statistical difference in the eradicative rates between Nature Chinese drugs and Nature Chinese drugs +double therapy (P>0.05). In the metronidazole susceptible group:The eradicative rates of Nature Chinese drugs (Laggera pterodonta+Pomegranate Rind+Taraxacum mongolicum ) % Nature Chinese drugs +double therapy(Colloidal bismuth subcitrate + tetracycline haydrochloride ) and triple therapy (Colloidal bismuth subcitrate+ metronidazole+tetracycline haydrochloride)on H.pylori is 66.7%% 80% and 86.7%. There is no statistical difference in the eradicative rates between Nature Chinese drugs+ double therapy and triple therapy (P>0. 05); There i s no statistical difference in the eradicative rates between Nature Chinese drugs and triple therapy(P>0.05); There is no statistical difference in the eradicative rates between Nature Chinese drags and Nature Chinese drugs +double therapy(P>0.05). In the different groups : The eradicative rates of triple therapy in the metronidazole susceptible group is better than in the metronidazole resistant groups,the difference has a statistical significance (P<0. 05); The eradicative rates of Nature Chinese drugs in the metronidazole susceptible and resistant groups have no statistical significance (P>0. 05); The eradicative rates of Nature Chinese drugs+double therapy in the metronidazole susceptible and resistant groups have no statistical significance (P>0.05) Conclusion: (1) H.pylori metronidazole susceptible strains and resistant strains weresatisfactorily susceptible to Laggera pterodonta> Pomegranate Rind and Taraxacum mongolicum./ï¿¡p>'/on metronidazole resistant strains and susceptible strains was suscetible to it when they are used singly. (2) Orthogonal Test Method filter the best combination of Natural Chinese drugs of anti-H.pylori metronidazole susceptible strains and resistant strains: Laggera pterodonta(0.278g/ml): Pomegranate Rind(0.960g/ml): Taraxacum mongolicum(0.137g/ml) and Laggera pterodonta(0.179g/ml): Pomegranate Rind(1.116g/ml): Taraxacum mongolicum(0.460g/ml).This test can enhance the efficiency to filter the best combination of three drugs and reduce the times of test and save the resource of scientific research. (3) This study find that the eradicative rates is significant higher in H.pylori metronidazole susceptible groups than H.pylori metronidazole resistant groups. The eradicative rates of Nature Chinese drugs+double therapy is better than trilp therapy to treat the metronidazole resistant strains infected animals. (4) Establish a metronidazole resistant strains and susceptible strains infection model of BALB/c mouse successfully with Yunnan strains: 1-139 and 1-145. The Yunnan strains 1-139 and 4-145 with CagA+ and VagA+ have the better effect of inoculation used in the study of H.pylori infected BALB/c mouse and is worth of generalized and applicated abroadly.The improved surgery can enhance the rate of inoculation and save the amount of bacterium, and have the better stability. PCR-RFLP is the best differential method which can identify the H.pylori in animal model. (5)The three Nature Chinese drugs has a abroad distribution and abundantly resource, it is cheap and has no medical resistant and side-effects, they can be used in the health care of H.pylori infection.The combination of Nature Chinese drugs can be used in the treatment of H.pylori infection and the remedy after H.pylori metronidazole resistance.Find a new anti-H.pylori drugs generalized and employed in city and country.There is a immeasuralbe social and economic values to the prevent and treatment of H.pylori related diseases. (6)At homeand aboard,the inhibitive effect of Laggera pterodonta on H. pylori in vitro has not been reported .There are no reports about the inhibitive effect of either Pomegranate Rind or Taraxacum mongolicum on H. pylori in vitro.
