Objective:1.Investigate the resistivity of Hp of gastric mucous membrane of patients with upper gastrointestinal disease to three commonly used antibiotics(metronidazole, charithromycin, amoxicillin), screen out resistant strains.2.To explore the inhibiting effects of total alkaloids of sophora alopecuroides (TASA),and monomer alkaloids(matrine,oxymatrine,and sophoridine)in vitro. Explore the feasibility of the therapy of Hp infection.Methods:1.To collect the specimens with rapid urease test positive from gastric mucous membrane of patients.Using Colombian agar medium,Helicobacter pylori was isolated, cultivated and identified Combined with morphology and biochemistry.2. Antimicrobial Susceptibility Test of42clinically isolated Helicobacter pylori strains are examined with E-test. And analyzes Hp to antibiotic resistance characteristic with different gender, age and type of disease.Total27Hp strains were filtered as resistant strains3.The minimal inhibitory concentration(MIC)of total alkaloids of sophora alopecuroides (TASA),matrine,oxymatrine,and sophoridine are examined by tube dilution method.Results:Get gastric mucosa specimens by gastroscope.Using Colombian agar medium,microaerophilic environment(5%O2,10%CO2å'Œ85%N2),37℃,humidity>95%.after cultiv-ation3-5days,they are visible to the naked eye,that on the surface of the medium colonies are pinpoint-sized,translucence,moist,and like dew.Smear dyeing and under the microscope,the bacterium was gram-negative,curved,spiral,look like a spread-winged seagull.And urea enzyme test,oxidase test,catalase test were positive.Can identified the isolated and cultivated strains were Helicobacter pylori.there are totally cultivated42Helicobacter pylori.2.After antimicrobial Susceptibility Test by E-test in vitro,27strains resistance to metronidazole and6strains to clarithromycin.The antibiotic resistance rates were64.3%and14.3%respectively.At the same time appear multiple drug-resistant strains.Has not yet detected Hp resistance to3. The50%minimal inhibitory concentration (MIC50) are found to be16mg/ml,64mg/ml,64mg/ml,128mg/ml for the Total alkaloids of sophora alopecuroides (TASA), the matrine, the oxymatrine, and the sophoridine, respectively.Conclusion:1.Since clinically isolated Hp is highly resistant to metronidazole,those metronidazole-contained medicines should not be selected as first-choice treatment for Hp infections.It has a high resistivity to clarithromycin; therefore, the latter should be used seletively with drug testing. HP is sensitive to amoxicillin; hence, the latter should be used as a major medicine for curing Hp infections.2. The50%minimal inhibitory concentration (MIC50) of the Total alkaloids of sophora alopecuroides (TASA) was16mg/ml.Considering the good performance of the total alkaloids of Sophora alopecuroides(TASA) in inhibiting Helicobacter pylori in vitro,TASA medicine would be an alternative to treat Helicobacter pylori infection.
