The formulations of rifapentine and isoniazid were studied in this work. Developed and characterized rifapentine self-emulsifying drug delivery systems (SEDDS) , and isoniazid matrix sustained-release capsule. These two formulations were optimized, and the properties of formulations were studied.Developed and characterized SEDDS of rifapentine, SEDDS formulations were prepared using different oils, surfactants and cosurfactants. In all the formulations, the in vitro self-emulsification properties and release of rifapentine were studied. From these studies, an optimized formulation consisting of 1.5mL OP10-Span85, 10g PEG4000, 10mL corn oil, 3g rifapentine was selected, there were 36 folds increase in the absorption for the self-emulsifying system compared to a common formulation.According to literature, UV spectrophotomatry was developed for assay of rifapentine during the studies of physicochemical properties, content and release. High-performance liquid chromatography was applied in in vitro simulated absorption and in formulation optimizing.On the basis of rifapentine SEDDS studies, the preparation technique and optimized formulation of isoniazid sustained-release capsule, which extend the drug release to 8 hours, were determined by orthogonal design test and uniform design according to cumulative percent of drug release. UV and HPLC methods were developed for assay during the studies of content and release. The load of sustained-release capsules was 320±20mg, including 100±8mg of isoniazid. The different models were used to study the releasing mechanism of isoniazid sustained-release capsules, which accorded with criterion of assay and FDA release limit.
