Objective: To explore the reason why neither morbidity nor serious degree of acute graft versus host disease (aGVHD) after allogeneic peripheral blood stem cell transplantation (APBSCT) are higher than that after allogeneic bone marrow transplantation (ABMT) by studying the impact on expression of lymphocyte function antigen-1(LFA-1), intercellular adhesion molecule (ICAM-1), L-Selectin(LAM-1) and very late antigen 4a(CD 49d) on CD4~+ T cells, and functions of CD4~+ T cells through these molecules during mobilization with recombinant human granulocyte colony stimulating factor (rhG-CSF). Methods: (1)The proportion between CD4~+T and CD8~+T cell, and the expression of LFA-1, CD54, CD49d and CD62L of peripheral blood CD4~+ T cells were determined by FACS before rhG-CSF mobilization and the fifth day of mobilization.(2) CD4~+ T cells were separated by MACS. The migration rate of CD4~+ T cells to SDF-1α and the adhesion activity of CD4~+T cells to ICAM-1 were tested separately before rhG-CSF mobilization and the fifth day of mobilization. (3)IFN-γ and IL-4 secreted by CD4~+ T cells were examined with ELISA. (4) IFN-γ, IL-4 and IL-10 mRNA expression of CD4~+T cell were detected with RT-PCR analysis.Results: (1)The the value of CD4~+/ CD8~+ was 1.35±0.27 and 1.30±0.29before mobilization and the fifth day of mobilization respectively, didn't change markedly, (P>0.10).The expression of LFA-1 on CD4~+T lymphocytes were both 100%. The expression of VLA-4(CD49d) was ( 79.17±13.64 ) % and ( 81.6±9.58) % respectively, didn't change markedly before and the fifth day of mobilization,(P>0.10). The expression of LAM-1(CD62L) on CD4~+ T lymphocytes was ( 50.79±21.7) % and (12.9±8.73 ) %,and the expression of ICAM-1(CD54) was (49.49±15.75 ) % and (14.76±8.12) % respectively, bothdecreased markedly during mobilization, (P<0.01).(2)SDF-la induced CD4+ T lymphocytes migration for 4 hours was (28.56±5.45 )% and( 31.35±5.33 )% respectively, didn't change markedly duringmobilization (P>0.10) .(3)The adhesion activity of CD4+T lymphocytes to ICAM-1 was( 84.4±6.45 ) % and ( 60.69±10.18 ) % respectively, decreased markedly during mobilization, (P>0.10) .(4) The concentration of IL-4 in peripheral blood serum was 21.15±7.73pg/ml and 12.65±6.43pg/ml, and the concentration of IFN-y was 43.03±18.05pg/ml and 20.74±7.08pg/ml respectively, both decreased markedly during mobilization,(P<0.01).Conclusions: rhG-CSF does not present effect on the value of CD4+/ CD8+,the expression of LFA-1 and VLA-4 on CD4+ T cell,and the migration activity of CD4+ T cell during mobilization,but can effect the expression of ICAM-1 and LAM-1 on CD4+ T cell,and the adhesion activity of CD4+ T cell to ICAM-1 through LFA-1,and maybe effect CD4+ T cell secretion function of IFN-y and IL-4.
