Objective: To investigate the influence of high selective Cyclooxygenase-2(COX-2) inhibitor celecoxib on proliferation and apoptosis of SK-N-SH cell and reveal the potential COX-2-independent mechanism of celecoxib effect on SK-N-SH.Methods: After SK-N-SH cells were treated with celecoxib in different doses( 12.5μumol/L, 25/μnol/L, 50μmol/L, 75μmol/L) and different time(24 hours,48 hours,72hours), Methabenzthiazuron (MTT) assay was used to measure proliferation . DNA ladder and AO/EB staining were used to observe the apoptosis. The expression of COX-2 and Bcl-2 were detected by Western blot before and after treatment with celecoxib.Results: Celecoxib inhibited the cell's proliferation in a dose- and time-dependent manner. The typical DNA ladder and the morphology of cell apoptosis can be observed through DNA elec-trophoresis and AO/EB staining. The expression of COX-2 and Bcl-2 were detected in SK-N-SH cells. The expression level of them decreased with the increase of the dose of celecoxib and the effect time .Conclusion: Celecoxib inhibits growth and induces apoptosis of SK-N-SH cells through down-regulating the expression of COX-2 and Bcl-2 in vitro. The COX-2-independent effect may exist. This provides a new therapeutic option for neuroblastoma patients.
