| Objective: To investigate the proliferative effect of rhEGF under different concentration and the abnormal differentiation effect of rhEGF under certain concentration on human keratinocyte growth. Methods: the standard medium was keratinocyte growth medium with FBS, hydrocort, insulin, and adenine. Keratinocytes from a 1.5-year-old subject were culture and seed on dishes with the standard medium in cell density of 1×10~5/25mm~2. After 7 days, the medium was added the standard medium with 0.001,0.01,0.1,1,10,100, and 500μg/L rhEGF, respectively. The standard medium without rhEGF was used as the negative controls. The growth of keratinocyte was monitored by 3- (4,5-dimethythiazol-2-yl)-2,5 dipheyl tetrazolium bromide (MTT) assay and by photographs on day 12, respectively. Results: rhEGF in concentrations of 0.01—100μg/L showed a significant proliferative effect on days 12 as compared with that of the negative controls (P<0.05), but less than 0.001 u g/L wasn't the proliferative effect with the same controls. With FCM,rhEGF in concentrations of 1 u g/L showed a normal poliferative effect. Conclusion: rhEGF has significant effects on the proliferative of human keratinocyte. The optimal response occurred when the concentration of rhEGF was 0.01—100μg/L.There is no abnormal proliferative change on that concentration. |
