| Objective To master the cell culture technology and the growth character of fibroblasts. To investigate the proliferative effect of rhbFGF , rhEGF under different concentration and the abnormal differentiation effect of two growth factors under certain concentration on human skin fibroblasts .Methods Interfere in the fibroblasts in vitro culture of rhbFGF , rhEGF under different concentration and detect the cell's process activity with the method of MTT. Select the cell under the best concentration of these two growth factors and detect their DNA ploid with FCM.Resultsâ‘ Observe fibroblasts process of growth: fibroblasts adherence on the whole after passage and others are gotten rid of in the process of changing culture liquid. After 4-5 days, the cell growth to 80 percent of culture flask and continue passage. The cell offer fusiform shape and have uniformity distribution.â‘¡The proliferation effect of rhbFGF under different concentration on fibroblast: MTT value shows 50ug/L as the powerful concentration.â‘¢The proliferation effect of rhEGF under different concentration on fibroblast: MTT value shows 80ug/L as the powerful concentration.â‘£Detect cell's DNA ploid with FCM: with FCM, both two growth factors in the powerful concentration showed normal proliferative effects.Conclusions rhbFGF and rhEGF all have significant effects on the proliferative of fibroblasts. The best response occurred when the rhbFGF concentration reach to 50ug/L while rhEGF concentration to 80ug/L. Compared with these two concentrations, rhEGF has bigger effects than rhbFGF. Utilizing optimal concentration will hasten the wound healing and we'd better pay attention to the importance of effective concentration. |
