Objective: To study of the expression and its clinical significance of c-erBb-2, nn23, PS2 and Ki-67 in breast cancer. Method: The expressions of c-erBb-2, nm23, PS2 and Ki-67 were detected by IHC and tissue chip from 278 cases of breast cancer. Results: (1)19 tissue microarrays were made by hand. Factures of tissue microarray were integrity, exquisite and no splits. The cores in tissue microarrays were arranged clearly. They were answered for the request of research. According rate of nm23 by immunohistochemical technique between the tissue chip and the whole tissue sections were 91.40% (85/93). There were no significant differences between them and the whole section (P>0.05). (2)The positive rate of c-erBb-2, nm23, PS2 and Ki-67 was 51.42%, 69.79%, 37.41% and 47.84%, respectively. (3)The expressions of c-erBb-2 and PS2 showed that they were positive correlation with axillary lymph metastasis and clinical stage, but the expression of nm23 was oppositely. At the same time, the expression of c-erBb-2 was associated with age and menopausal status. The expression of nm23 was negatively and related to the tumor size. (4)The expressions of c-erBb-2 and PS2 were associated with the expressions of ER, PR and ER-PR. The expressions of c-erBb-2, nm23 and PS2 were relatively.Conclusion: (l)Tissue chip was a new technique which has characters of high-throughout, economy, and low rate of experimental error, and it was better used on large sample and retrospective clinical research. (2)The method of tissue chip made by hand was simple, reliability and economical technique, and could be used on clinicopathological diagnosis and research work. (3)The cores in tissue chip were representative, and could reflected the informations of the whole-tissue. When mading tissue microarray, taking 23 cores from the whole-tissue sections were an accurate and economical method. (4) The expressions of c-erBb-2, nni23 and PS2 were related to progression, invasiveness and metastasis with breast cancer. And both c-erBb-2 and PS2 gene expression correlated with ER, PR. (5)Detectionofc-erBb-2, nni23? PS2andER, PR, these would be better to evaluate malignancy and metastasis especially in axillary node negative breast cancer(ANNBC), and would be helpful in screening the high risk cases for further treatment. (6)C-erBb-2, nni23 and PS2 were significance prognostic indices of breast cancer.
