Objective To observe the renal protection of mycophenolate mofetil in type 2 diabetic rats and to investigate the mechanism.Methods Forty-eight Wistar rats(female,160-180g body weight)were divided into three groups: normal control rats,diabetic rats,diabetic rats treated with mycophenolate mofetil (MMF, 15mg.kg-1.d-1) . Blood glucose,blood insulin,blood lipid,urinary albumin excretory rate and creatinine clearance rate were measured after 13 weeks.Kidney pathology was observed. Inmmunohistochemistry was used to analyse the expression of CTGF,MMP-9,TIMP-1 and MCP-1.Results No significant differences of blood glucose and blood insulin were found between diabetic rats and mycophenolate mofetil treatment group. Compared with the untreated diabetic group, urinary albumin excretion rate and creatinine clearance rate of MMF treated rats were decreased significantly .The expression of CTGF,TIMP-1 and MCP-1 in MMF group were much lower than diabetic untreated group,while the expression of MMP-9 was upregulated. MMF also improved the histological changes of diabetic rats.Conclusion mycophenolate mofetil has some renal protective effect on type 2 diabetic rats,partly through down-regulating CTGF and MCP-1 expression,regulating the balance of MMP-9 and TIMP-1 and reducing monocyte/macrophage infitration.
