Objective: 1.To investigate the influence of different blood purifies on the serum leptin levels(SLL) in the patients with chronic renal failure. 2.To explore the expression of cogenes: c-myc, c-fos on the kidney of SHR..3. To explore the protection of Valsartan on the kidney of SHRMethods: Sixty patients with maintenance hemodialysis in our hospital were included,which were divided into 3 sets:HD set,HF set, HP set,and SLL were measured by radioimmunoassay(RIA) before and after treatment.Twenty-four SHRs with the age of 55 weeks old and 12 WKY with the same age were involved. Bisexual and each half.They were divided into 3 groups: 1 .WKY-control group,which were fed distilled with water on time per day;2.SHR-control group,which were fed with distilled water on time per day;3.SHR-Valsartan group,which were fed with Valsartan(20mg/kg/d). After 12 weeks treatment,all rats were drawn for detection,including (1)the level of PRA , AngII and ALD. (2) the m RNA expression levels of c-myc and c-fos on kidney by RT-PCR.(3)the protein levels of c-fos and c-myc were detected by the immunohistochemical methods.Results: 1. SLL in MHD were obviously higher than the healthy controls and hemodialysis has little effect on SLL, while HF and HP can lower SLL significantly and HP surpasses HF.2. .Systolic blood pressure(SBP) after 12 weeks treatmentThe value of SBP in SHR-control group was significantly higher than that of the same age-group WKY group(p<0.01) SBP value of Valsartan group was significantly lower than that in SHR-control group(p<0.01)3. Blood serum hormones detectionPRA of both SHR groups are not significantly difference(P>0.05),but higher than that of the WKY group.. Angll of SHR-control group was significantly lower than that of the WKY group(P<0.05),while that of the valsartan group was much higher.ALD of the SHR-control group was higher than that of WKY group(P<0.01),while that of valsartan group was significantly lower(P<0.01).Conclusions: HP and HF can eliminate SLL preferably,which may improve the patients' nutritional status. The gene and protein expression of c-myc and c-fos in renal tissue of SHR enchances significantly, and may play an important role in the renal injury of hypertension. Valsartan amends renal injury by downregulating the gene and protein expression of c-myc and c-fos through blocking the tape I receptor of Angll.
