BackgroundRenal ischemia-reperfusion injury is not only an invariable consequence of transplantion but also results from shock and complicated operation.Recently,in the wake of the study of free radical biomedicine,biochemistry disorder and tissue disorganization results from free radical are considered to be etiopathogenesis.A free radical is any species capable of independent existence that possesses one or more unpaired electrons and thus is chemically highly reactivity.Oxygen free radicals include superoxide radicals hydroxyl radicals and hydrogen peroxide and rare exist in normal organism because superoxide dismutase (SOD) catalase(CAT) GSH-px in histiocytes eliminate them immediately.Massive oxyradicals are produced byvascular endothelial cells and neutrophils after renal ischemia-reperfusion injury and accumulate in renal because of the dysfuction of SOD CAT GSH-px resulting fron ischemia and hypoxia. Oxyradicals have powerful activity of oxidation and hazard almost all kinds of biomolecules by peroxidation.Biological membrane(include cellular membrane,mitochondrial membrane,lysosomal membrane,etc)contain aplenty polyunsaturated fatty acid(PUFA) and are prone to be attacked by oxyradicals to produce dead end products ,such as MDA.Dysfuction of biomembranes due to disproportion of polyunsaturated fatty acid and protein aggravate renal ischemia- reperfusion injury.Edaravone is a potent and novel scavenger of free radicals inhibiting hydroxyl radicals but also free radicals mediated lipid peroxidative damage.It is extensively applicated in patients with acute cerebral infarction because of its protective effects on cerebral edema and tissue injury after ischaemia-reperfusion injury.However,application of edaravone in renal ischemia-reperfusion injury has rare been reported.We applied edaravone injection to the rats with acute renal ischemia-reperfusion injury model to study the protective effects of edaravone on ischemia-reperfusion injury.Animal experimentObjective To investigate the effect of edaravone on ischemia-reperflision injury of rat renal.Methods The rats were divided into three groupsigroup A(sham operation),group B (renal ischemia-reperfusion) and group C (edaravone treated).Serum maleic dialdehyde(MDA),superoxide dismutase(SOD), blood urea nitrogen(BUN),creatinine(Cr), renal MDA and SOD were measured after 24h reperfusion.Renal ultrastructure were observed.Results Serum MDA(4.35±0.78nmol/L),renal MDA (15.03±1.19nmol/mgprot),serum Cr(170.9±16.1umol/L) of group C were significant lower(P<0.01) than those group B: Serum MDA (7.70±2.17nmol/L), renal MDA(19.74±1.71 nmol/mgprot), serum Cr( 233.2±17.2 umol/L). Serum SOD(355.2±40.2U/ml),renal SOD (26.02±7.75U/mgprot) of group C were significant higher(P<0.01) than those in group B:semm SOD(253.3±42.0U/ml),renal SOD(10.26±1.03 U/mgprot).Compared with group B3UN level of group C was no significant changes(P>0.05).The renal ultrastructure was greatly injured in group B,meanwhile,it was obviously ameliorated in group C.ConclusionK Edaravone can eliminate oxyradicals effectively after renalischemia-reperfiision injury. 2^ Edaravone can relieve injury of renal ultrastructure after renalischemia-reperfiision. 3^ Edaravone has protective effects on ischemia-reperfiisioninjury of rat renal.
