IntroductionMyasthenia gravis (MG) has immune dependence of cell, the immunity of body fluid mediated, the complement is participated in, to neuromuscular junction postsynaptic membrane, anti - skeletal muscle nicotine acetylcholine receptor antibodies, selfsensitization and autoimmune disease due to destruction. IFN -γ is secreted by Th1 cell, can induce B cell ripe, assist the production on AchR antibody, and induce the production of clinical symptoms. So may has some relation with MG origin. TGF - β1 can inhibit multiplication and division of B and C cell;can inhibit secret of Th1,Th2 cytokine;can participate in negative control response of cytotoxicity T cell related antigen 4( CTLA4) in cell activation incooperation simulate access, is main effector of immunotolerance. Thymus is main organ to induce T cell division and ripe, thymus structure and function is abnormal, may lead to autoimmune and autoimmune disease.Now immunology research of myasthenia gravis has made some process, but it's nosogenesis still didn't be found out, this test adopted immunohistochemistry to inspect TGF -β and IFN -γ representation in thymoma in Patients with Myasthenia Gravis to further discuss nosogenesis of Thymic Tumors in Patients with Myasthenia Gravis. These provide theoretical foundation for clinical MG patients immunization therapy, and provide research foundation for clinical application pharmacology method addition body TGF - β1 synthetic amount and activation and decreasing the production of IFN - γ.Materials and Methods1. Source of tissue sample.Specimen came from thoracic surgery of China Medicine University No. 1 attached hospital during from Jan. 1992 to June 2004. Thymoma specimen cut in operation are 72 cases, among them thymoma in Patients with Myasthenia Gravis were 48 cases, thymoma in patients without myasthenia gravis were 24 cases. Patients included 24 cases male and 48 cases female. Span of age is between 22 to 63 , average age is 46. 36 ± 1. 81. According to improved Osserman, this team MG patients were divided: I type (eye muscle type) 16 cases, Ila type (low grade whole - length type ) 14 cases, lib type ( moderate whole - length type) 16 cases, III type (severe active type)O case, IV type (hang-fire severe type)2 cases. All patients have not been receipted by radiate therapy and chemo - treatment.2. Immunohistochemistry.Paraffin embedding, 5 jjim slice, boast slice for 5 hours, put routine de -wax into water, tissue antigen microwave restoration, deactivation tissue peroxide enzymatic activity, seal up 5 ~ 10 percent of normal goal blood serum (PBS dilution) ,rabbit antihuman globulin TGF - (31 antibody of 1" 100 dilution, water bath 37°C for 3 hours, IFN - y antibody of 1 '■ 100 dilution, neutral balata mounting, DAB chromogenic for 4—5 min, haematoxylin counterstain, dehydration.Result1. TGF - pi representation level in thymoma in patients with myasthenia gravis is higher than thymoma in patients without myasthenia gravis ( t = 2.2380583 > t0 05/2(70) = 1.994 ,P <0.05) o2. IFN - y representation level in thymoma in patients with myasthenia gravis is higher than thymoma in patients without myasthenia gravis ( t = 2.11602767 >t005/2(70) = 1.994, P <0.05) o3. Representation level of TGF - pi and IFN - 7 MG patients has no relation with sex and age, and with I, Ha, lib , IVof Osserman.DiscussionMyasthenia gravis is autoimmune disease to neuromuscular junction AchR, the original area is uncertain, but now think that it is possible in thymus, patients with thymoma often appear immune system imbalance, so induce autoimmune disease.Interferon - y( IFN - 7) is secreted by Thl cell, can induce B cell ripe, to assist the production on AchR antibody, and to induce the production of clinical symptoms. This experiment results show that IFN - 7 representation level in thymoma in patients with myasthenia gravis is higher than thymoma in patients without myasthenia gravis. There is statistical discrepancy, explain that IFN — 7 has close relation with MB onset, rising up IFN - 7 will promote MG occur. This experiment also show that MG patients Thymoma organ IFN - 7 level have no relation with sex, age and Osserman, explain that sex, age and Osserman is not decisive factor to influence IFN - 7 level.TGF - (31 is regarded as important endogenous immune rejection capability cell factor, play an important role at negative feedback regulation of disease immune. This experiment results show that TGF - pi representation level in thymoma in patients with myasthenia gravis is higher than thymoma in patients without myasthenia gravis. There is statistical discrepancy, explain that TGF - (31 has close relation with MB onset, MG patients immune function is imbalance, at the same time of starting immune activation mechanism, also start immune inhibit function. This experiment also show that MG patients thymoma organ TGF - pi level has no relation with sex, age and Osserman, explain that sex, age and Osserman is not decisive factor to influence TGF — pi level.In brief, in the process of acute stage MG onset, immune regulation function disorder, plus -minus immune regulation process all be activated. IFN -7 may has some relation with disease origin. TGF - pi plays important role in inhibiting MG onset, make it become a kind of possible cure method to cure MG,so it has brighter prospects.ConclusionIn conclusion, we have shown here that IFN - "y and TGF - (31 are expressed in thymoma in patients with myasthenia gravis is higher than thymoma in patients without myasthenia gravis, both of them have the significance of statistics. It shows that IFN - *y and TGF — JBl have an important significance in thymoma in patients with myasthenia gravis. IFN - y has function that promotes the occurrence and development of thymoma in patients with myasthenia gravis, TGF - pi has function that restrains the occurrence and development of thymoma in patients with myasthenia gravis.
