Objective: To observed the change of PGE2 and angiopoietin-l(Angl) and angiopoietin-2(Ang2) protein in human tongue squamous cell carcinoma Tca8113 cell lines by inhibiting activated COX-2 for investigating potential mechanism of COX-2 and the selective COX-2 inhibitor nimesulide(NIM) affect biological behavior of human tongue squamous cell carcinoma. Method: Tca8113 cell were cultured in vitro,and exposed to a range of concentrations of nimesulide(NIM)(0,25,50,100,200μmol/L)for 24h,48h,72h,and the PGE2 level in the cells cultured supernatant was quantitated by the radioimmunoassay, and the expression of Angl and Ang2 protein in human tongue squamous cell line Tca8113 Were measured by the SABC immunohistochemistry staining simultaneously.Results: Tca8113 cell were treated with NIM(25,50,100,200μmol/L)for 24h 48h 72h ,and (1 )the comparison of level of PGE2 and expression of Ang2 protein in the group (P<0.05) and time (P<0.05) was statistical significance, but not Angl in the group (P>0.05) and time (P>0.05) .(2) The variational tendency of PGE2 and Angl was not correlation, r=—0.364, P>0.05;The variational tendency of PGE2 and Ang2 was correlation, the correlation coefficient of PGE2 and Ang2 was 0.579, PO.05.Conclusion:The PGE2 level in the cultured supernatant and Expression of Ang2 decreased by NIM in a concentration and time dependent manner,but not Angl. The variational tendency of PGE2 and Angl was not significant correlation.The variational tendency of PGE2 and Ang2 was positive correlation.NIM can inhibit Cox-2 activity and down-regulate PGE2 and Ang2 expression, which may be effected on Cox-2 by the inhibition of angiogenesis and involved in cancer inhibition mechanisms of Cox-2 inhibitor.
