Objective:Through observing the change of COX-2,vascular endothelial growth factor (VEGF),angipoietin-1(Ang-1)and angiopoietin-2(Ang-2)gene in human tongue squamous cell carcinoma Tca-8113 cell line by inhibiting the activity of COX-2 to investigate the affect and mechanism of the selective COX-2 inhibitor nimesulide(NIM)in the biological behavior of human tongue squamous cell carcinoma.Method:Human tongue squamous cell carcinoma Tca-8113 cell were cultured in vitro and treated with NIM(25,50,100,200μmol/L)for 48h to measure the expression of COX-2,VEGF,Ang-1 and Ang-2 mRNA in Tca-8113 cells by the semiquantitative reverse transcription polymerase chain reaction(RT-PCR)analysis.Results:Human tongue squamous cell carcinoma Tca-8113 cell were treated with NIM(25,50,100,200μmol/L)for 48h,the expression of COX-2 mRNA level between the each treated group and the control group was statistical significance(P<0.01),The expression of COX-2 mRNA reduced in a concentration dependent manner r=-0.963;VEGF mRNA level between the each treated group and the control group was statistical significance(P<0.01),The expression of VEGF mRNA reduced in a concentration dependent manner r=-0.934;Ang-1 mRNA level between the each treated group and the control group was statistical significance(P<0.01),The expression of Ang-1 mRNA reduced in a concentration dependent manner r=-0.988;Ang-2 mRNA level between the each treated group and the control group was statistical significance (P<0.01),The expression of Ang-2 mRNA reduced in a concentration dependent manner r= -0.954.Conclusion:Human tongue squamous cell carcinoma Tca-8113 cell were treated with NIM(25,50,100,200μmol/L),,the expression of COX-2,VEGF,Ang-1 and Ang-2 mRNA level in the each treated group compared with the control group was reduced,and the reduced in a concentration dependent manner.COX-2 inhibitor nimesulide(NIM)effected on the COX-2 to inhibit the expression of VEGF,Ang-1 and Ang-2 gene.The inhibition of angiogenesis in cancer can provide theoretical foundation for the COX-2 selective inhibitor nimesulide(NIM)in chemopreventive strategy of the cancer.
